摘 要:目的观察藏药砂生槐和阿苯达唑单独及联合用药治疗小鼠继发性肝包虫病的效果。方法 74只雌性昆明小鼠,除10只用生理盐水作空白对照,其余感染小鼠分为6组,分别为砂生槐高[16mg/(kg·d)]、中[12mg/(kg·d)]、低[8mg/(kg·d)]剂量组,阿苯达唑组20mg/(kg·d),砂生槐+阿苯达唑[8mg/(kg·d)+10mg/(kg·d)]及模型对照组。用原头节稀释混悬液腹腔注射构建实验动物模型,药物治疗3个月后,采血,检测小鼠血清IL-2、IL-6、IL-10、IgE和TNF-α、AST及ALT的含量;解剖小鼠,取脾、胸腺及包囊,称重,计算胸腺指数、脾脏指数及抑囊率。结果治疗3个月后,小鼠血清IL-2、IL-10、IgE含量及囊重联合用药组与模型组相比差异有统计学意义(P<0.05),AST及ALT联合用药组较空白对照组的增幅小于模型组增幅。病理显示各组脾脏组织中均出现单核或多核的巨细胞,阿苯达唑治疗组脾脏和肝脏均出现沉积物,其他治疗组(包括联合组)均无沉积物。结论砂生槐联合阿苯达唑治疗小鼠包虫病有一定效果,小鼠免疫力提高。同时,联合用药可减轻药物的肝毒性。Objective To observe the efficacy of Sophora moorcroftiana or albendazole alone or in combination in treating secondary hepatic hydatid disease in mice. Methods A total of 74 female Kunming mice were divided into groups:10 mice were treated with saline as a blank control and the remaining mice were divided into 6 groups:a high-dose Sophora moorcroftiana group[16 mg/(kg·d)],a medium dose group[12 mg/(kg·d)],a low dose group[8 mg/(kg·d)],an albendazole group[20 mg/(kg·d)],a Sophora moorcroftiana+albendazole group [8 mg/(kg·d)+10 mg/(kg·d)],and a model control group.An experimental animal model was created via intraperitoneal injection of a diluted suspension of protoscoleces.After three months of drug treatment,blood samples were collected from mice to determine serum IL-2,IL-6,IL-10,and IgE and levels of TNF-α,AST,and ALT.The mice were dissected,and the spleen,the thymus,and cysts were removed and weighed.The thymus index,spleen index,and inhibition rate were calculated. Results After 3 months of treatment,the levels of IL-2,IL-10,and IgE and cyst weight in mice in the Sophora moorcroftiana+albendazole group differed significantly from those in the model group(P 〈0.05).The increase in levels of AST and ALT in the Sophora moorcroftiana+albendazole group was smaller than that in the model group.A pathological examination revealed mononuclear or multinucleated giant cells in the spleen of each group.Sediment was found in the spleen and liver of the albendazole treatment group.No sediment was found in the other treatment groups(including the Sophora moorcroftiana+albendazole group). Conclusion Sophora moorcroftiana combined with albendazole has a certain level of efficacy in treating secondary hepatic hydatid disease in mice,and this combined treatment can improve the immunity of mice.Combined use of drugs can reduce the liver toxicity of a drug.
