复方诃子口服液治疗犊牛大肠杆菌性腹泻的研究

摘　　要：为了研究治疗犊牛大肠杆菌性腹泻的中药制剂,依据大肠杆菌病的中兽医辨证理论进行组方并制备复方诃子口服液,采用牛津杯法及试管二倍稀释法检测复方诃子口服液的抗菌活性、最小抑菌浓度（MIC）和最小杀菌浓度（MBC）;采用人工感染C57BL小鼠制备腹泻模型,并将50只小鼠分为空白组,模型组,口服液高、中、低剂量组,通过口服治疗观察临床效果,检测体重及采食量的变化,并取肠道黏膜组织进行病理学观察。结果表明：复方诃子口服液浓度为0.25 g/mL时抑菌效果为中度敏感,浓度为0.5 g/mL、1.0 g/mL时抑菌效果为高度敏感,MIC为31.25 mg/mL,MBC为125 mg/mL。灌服复方诃子口服液后,各组小鼠腹泻症状均有所改善。给药后第5天,口服液各剂量组小鼠体重及采食量基本恢复正常,而模型组仍低于造模前水平;口服液高、中、低剂量组肠绒毛基本恢复正常,肠腺变性消失,有增生;而模型组小肠绒毛病变有所好转,但肠腺部分增生。说明复方诃子口服液对大肠杆菌具有抑制作用,对大肠杆菌性腹泻具有治疗作用。The aim of the present study was to investigate the treatment of calf Escherichia coli（E. coli） diarrhea in traditional Chinese medicine preparations. The prescription was constituted and the compound Myrobalan oral liquid was prepared according to the veterinary syndrome of E.coli disease. The antimicrobial activity,MIC and MBC of the compound Myrobalan oral liquid were measured by Oxford cup method and double dilution method. The diarrhea model was established by artificial infection with C57 BL mice in vivo test. 50 mice were randomly divided into blank group,model group and oral liquid high,medium and low dose groups. The clinical effect,body weight and feed intake were detected by oral treatment,and the intestinal mucosal tissues were collected for pathological observation. The results showed that the bacteriostatic effect was moderately sensitive when concentration of compound Myrobalan oral solution was 0. 25 g/mL. When the concentration was 0. 5 g/mL and1. 0 g/mL,the bacteriostatic effect was highly sensitive. The MIC was 31. 25 mg/mL,and MBC was 125 mg/mL. The symptoms of diarrhea in mice were improved after feeding the compound Myrobalan oral liquid. On day 5 after administration,the weight and feed intake of each oral solution group had been returned to normal,while the model group was still lower than that before. The shape of the intestinal villi basically returned to normal,intestinal degeneration disappeared,but the hyperplasia appeared in oral liquid high,medium and low dose group. The small intestine villi lesions of model group had been improved,but the intestinal glands were still hyperplasia. The results indicated that the compound Myrobalan oral liquid had an inhibitory effect on E. coli and had therapeutic effect on E. coli diarrhea.
