摘 要:目的建立测定硫酸羟氯喹原料药中苯酚含量的高效液相色谱(HPLC)法。方法采用高效液相色谱法,Inertsil C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(70∶30,V/V),流速1.2 mL·min^(-1),检测波长为270 nm。结果在上述色谱条件下,苯酚质量浓度在0.11~1.15μg·mL^(-1)范围内呈良好的线性关系(r=0.999 8),回归方程:Y=15.424 8X+0.003 7,平均回收率为99.9%(n=9)。结论实验证明该法操作简单、准确性高且专属性强,适用于硫酸羟氯喹原料药中苯酚含量的测定。OBJECTIVE To establish a method for the determination of phenol in hydroxychloroquine sulfate by HPLC. METHODS The sample was analyzed on an Inertsil C18( 4.6 mm×250 mm,5 μm) column with the mobile phase of acetonitrile-water( 70∶30,V/V); the flow rate was 1.2 mL·min(-1),and the detection wavelength was 270 nm. RESULTS Under the above HPLC conditions,the linear range of the phenol concentration was 0.11-1.15 μg·mL(-1)( r=0.999 8).The average recovery of phenol was 99.9%( n=9). CONCLUSION This method is convenient and fast for the quantitative determination of phenol in hydroxychloroquine sulfate.
