摘 要:通过对脱落酸(ABA)前处理方法进行优化,并采用高效液相色谱法测定了苜蓿根系样品中ABA的含量。首先,分别对提取溶剂、提取时间、料液比、提取次数进行条件优化,并建立了交联聚乙烯吡咯烷酮(PVPP)和SPE联用的净化方法。结果显示,最佳的前处理方法为提取溶剂80%甲醇,提取时间16h,料液比为1∶20g/mL,提取2次。通过方法学考察,ABA在0.05~1mg/L范围内线性关系良好,检出限为0.02μg/g,定量限为0.04μg/g。该方法稳定性、精密度、重现性良好,加标回收率为91.7~105.2%,可以应用于植物中ABA含量的测定。The optimal extraction conditions of ABA were explored,and the content of ABA in the root of Medicago sativacv.was determined by high performance liquid chromatography.Effects of extraction reagent and extract time,material-liquid ratio and extraction times on extraction yield of ABA were investigated,and a purification method based on the combination of PVPP and SPE was established.The results showed that the optimal extraction conditions were as follows:extraction time 16 h;material-liquid ratio 1:20 g/mL;twice extraction with 80% methanol.The method was of good stability,precision and reproducibility,and the recoveries of ABA in this experiment were in the range of 91.7-105.2%.It can be applied to the determination of ABA content in plants.
