摘 要:目的研究降脂宁有效部位最佳提取纯化工艺,为降脂宁的进一步研究和开发利用提供依据。方法采用比色法测定总黄酮、总蒽醌和总生物碱的含量,用HPLC法测定2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量,正交实验设计优选最佳提取工艺,单因素考察法筛选降脂宁有效部位大孔吸附树脂纯化工艺。结果降脂宁有效部位的最佳提取工艺:药材用10倍量体积分数为70%的乙醇回流提取3次,每次1.5h;降脂宁有效部位的最佳纯化工艺为:X-5型大孔吸附树脂,上样液质量浓度为0.1g·mL-1,树脂柱径高比为1∶9,除杂溶剂为5BV水,洗脱溶剂为体积分数50%乙醇,洗脱体积7BV,洗脱流速为4BV·h-1。结论该工艺操作简单、稳定可行、重复性较好,可用于降脂宁有效部位的制备。Objective To optimize the extraction and purification process of the effective fraction of Jiangzhining and provide reference for the further research,development and utilization of Jiangzhining.Methods The content of total flavones,total anthraquinones and total alkaloids were measured by using colorimetric method.And the content of 2,3,5,4′-hydroxyphenyl-vinyl-2-O-β-Dglucoside was determined by HPLC.The optimal extraction process was studied by applying orthogonal design test,and the purification process with macro-porous absorptive resins was screened by single factor test.Results The parameter of the optimal extraction process was as follows:70%ethanol(10 times of crude Jiangzhining)for extracting for 3 times and 1.5 hours each time.The optimal purification process was as follows:X-5 macro-porous absorptive resin,sample concentration 0.1 g·mL^-1,ratio of column diameter to height 1∶9,purification solvent water(5 BV),elution solvent 50% ethanol,elution volume 7 BV and elution flow rate 4 BV·h^-1.Conclusion The optimized extraction and purification process are stable and feasible,and could be applied to the preparation of the effective fraction of Jiangzhining.
