高效液相色谱法测定羟基喜树碱在不同有机溶剂和增溶剂中的平衡溶解度

摘　　要：目的测定羟基喜树碱(HCPT)在不同有机溶剂和增溶剂中的平衡溶解度,为该药新剂型的研发奠定基础。方法采用高效液相色谱法测定HCPT在不同有机溶剂和增溶剂中的平衡溶解度。结果确定了HCPT的高效液相色谱色谱条件,在9种常用有机溶剂中,N,N-二甲基甲酰胺对HCPT的平衡溶解度最高[(1 674.477±0.032)μg/m L];在7种常用增溶剂中,1%十二烷基硫酸钠溶液对HCPT的平衡溶解度最高[(60.853±0.016)μg/m L]。结论高效液相色谱法简单、可行,实验结果可为HCPT新剂型的研发提供科学依据。Objective To determine equilibrium solubilities of hydroxycamptothecin（HCPT）in different organic sol-vents and solubilizer agents so as to lay the foundation for the research and development of the new dosage form of the drug.Methods The high performance liquid chromatography（HPLC）method was established to detect the equilibrium solubility ofHCPT in various of organic solvents and solubilizers. Results The HPLC chromatographic conditions of HCPT were deter-mined.The equilibrium solubilities of HCPT was [（1 674.477±0.032）μg/m L] in N,N-dimethylformamide,the largest amongnight of common organic solvents;the equilibrium solubilities of HCPT was [（60.853±0.016）μg/m L] in 1% twelve alkyl sodiumsulfate solution,the largest among seven of common solubilizers. Conclusion HPLC method established in the study is simpleand feasible.The results in the study provides a scientific reference for studing and developing of HCPT′s new dosage form.
