摘  要:为研究安徽省日本沼虾(Macrobrachium nipponense)群体的遗传多样性,利用微卫星标记技术,采用10对微卫星引物对安徽省6个野生群体(女山湖NSH、巢湖CH、姑溪河GXH、南漪湖NYH、长江东至段CJ、升金湖SJH)和4个养殖群体(滁州CZ、太湖1号F2TH1H、当涂DT、养贤YX)的遗传多样性和遗传结构进行了分析。结果显示:所选用的10对微卫星标记均表现为高度多态性(PIC>0.5),各群体均具有较高的遗传多样性水平(期望杂合度He=0.795~0.876)。分子方差分析(AMOVA)显示,遗传变异有3.64%来自群体之间,96.36%来自于群体内部。遗传分化和遗传距离分析显示,群体间遗传分化属于中低水平(Fst<0.15),NSH和TH1H之间的遗传分化最小(Fst=0.002 8),遗传距离最近(D=0.039 8),而SJH和CZ之间的遗传分化最大(Fst=0.114 4),遗传距离最远(D=0.226 0)。基于群体间Nei’s遗传距离构建的UPGMA系统进化树显示,NSH和TH1H遗传关系最近。综上所述,10个群体遗传多样性水平较高,各群体间遗传分化属于中低水平。Microsatellite markers experimental technique was used in this research to study the genetic diversity- of Macro- brachium nipponense in Anhui Province. The genetic diversity- and genetic structure of 6 wild populations ( NSH, CH, GXH, NYH, CJ, SJH) and 4 cultivated populations ( CZ,TH1H, DT, YX) in Anhui province were analyzed by 10 pairs of mi- crosatellite markers. The results showed that high degree of polymorphisms were observed in all the selected ten nficrosatel- lite markers (PIG 〉 0. 5 ). Meanwhile, the high genetic diversity- was shown among all the populations ( He = 0. 795 - 0. 876 ). The AMOVA showed that 3.64% of genetic variation was from populations and 96.36% was from individuals. The genetic differentiation index (Fst) within populations was relatively low (Fst 〈 0. 15 ). Fst between NSH and TH1H was the lowest at 0. 002 8, while that between SJH and CZ was the highest at 0. 114 4. The genetic distance between NSH and TH1H population was the lowest at 0. 039 8, while that between SJH and CZ population was the highest, 0. 226 0. The UPGMA phylogenetie tree was constructed based on Nei' s genetic distance showed that the relationship between NSH and TH1H was the closest. All the results above showed that the ten populations had high levels of genetic diversity, the genetic differentiation among populations was in low or medimn level.
