摘 要:[目的]分析西藏牛和日喀则驼峰牛的Y染色体遗传多样性及父系起源。[方法]采用PCR扩增、测序及生物信息学方法。[结果]通过对13头西藏牛Y-SNPs分析,发现西藏牛包含普通牛Y1、Y2及瘤牛Y3三种单倍型组,其频率分别为0.077、0.846和0.077;对8头日喀则驼峰牛Y-SNPs的分析表明,日喀则驼峰牛具有普通牛Y2和瘤牛Y3两种单倍型组,其频率分别为0.125和0.875。西藏牛和日喀则驼峰牛的单倍型多样度分别为0.2949±0.1558和0.2500±0.1802,表明西藏牛和日喀则驼峰牛具有较低的父系遗传多样性。[结论]西藏牛主要为普通牛Y2起源,日喀则驼峰牛主要为瘤牛Y3起源,且其遗传多样性较低。[Objective] The aim of this study was to analyze the paternal origin and genetic diversity of Tibetan and Shigatse Humped cattle. [Methods] Polymerase chain reaction amplification, sequencing and bioinformatics methods were used. [Results] Based on the analysis of the Y-SNPs of 13 Tibetan cattle and 8 Shigatse Humped cattle, the results indicated that Tibetan cattle had three haplogroups, including Y1 and Y2 haplogroups of taurine and zebu Y3 haplogroup with frequencies 0. 077, 0. 846 and 0. 077. Moreover, Shigatse Humped cattle had taurine Y2 haplogroup and zebu Y3 haplogroup with frequencies 0. 125 and 0. 875. The baplotype diversities of Tibetan cattle and Shigatse Humped cattle were 0. 2949±0. 1558 and 0. 2500±0. 1802 respectively, suggesting that Tibetan cattle and Shigatse Humped cattle have relatively lower paternal genetic diversity. [Conclusion] Tibetan cattle mainly originated from taurine Y2, while Shigatse Humped cattle mainly originated from zebu Y3. Furthermore, in Tibetan cattle and Shigatse Humped cattle, the genetic diversity were low.
