摘  要:目的研究红树林植物红树、拉关木、秋茄树、海桑、玉蕊、阔苞菊的ITS2(DNA内转录间隔区2)条码鉴定方法,为红树植物药用价值的开发和优良种质资源的引进提供鉴定基础。方法对样本进行DNA提取、PCR扩增和双向测序。用CodonCode Aligner软件进行序列拼接,通过MEGA5.1软件比较各序列间的变异性,计算遗传距离;采用邻接法构建NJ(近邻结合法,Neighbor-Joining)系统树。结果 6种红树林植物的ITS2序列间存在明显变异,这种差异不仅体现在序列的碱基种类和数量上,更体现在ITS2序列遗传变异上。结论ITS2序列可准确有效地鉴别不同红树植物。Objective To study ITS2(Internal Transcribed Spacer 2)sequences identification method of mangrove plants Rhizophora apiculate Bl.,Laguncularia racemosa.Gaertn.C.F.,Kandelia candel Druce.,Sonneratia caseolaris(L.)Engl.,Barringtonia racemosa(Linn.)Spreng and Pluchea indica Less.,to provide the identification basis of medicinal value development and fine germplasm resources introduction.Methods After extracting the sample for DNA,amplifying PCR and employing bi-directional sequencing,the ITS2 sequences of the six mangrove plants were analyzed by using CodonCode Aligner for assembling,MEGA5 for genetic distance and constructing NJ(Neighbor-Joining)tree.Results ITS2 of the six mangrove plants showed variation not only on base type and quantity of sequences,but also on genetic variation.ConclusionITS2 sequences could be utilized to identify different mangrove species effectively and accurately.
