摘 要:目的研究清肠温中方对葡聚糖硫酸钠(DSS)诱导慢性溃疡性结肠炎(UC)模型小鼠Th1/Th2平衡的干预作用。方法 90只雄性BALB/c小鼠随机分为6组,除空白组外其余均自由饮用3%DSS溶液7d,之后换蒸馏水饮用7、14d为1个周期,共3个周期复制模型。用蒸馏水、清肠温中方高、中、低剂量和美沙拉嗪缓释颗粒灌胃干预14d,记录疾病活动指数(DAI),酶联免疫吸附法检测血清白介素-1β(IL-1β)、白介素-4(IL-4)、白介素-5(IL-5)、白介素-6(IL-6)及肿瘤坏死因子-α(TNF-α),免疫组化染色检测结肠黏膜NF-κBp50表达。结果与空白组比较,模型组小鼠DAI评分明显升高(P<0.01),血清IL-1β、IL-6及TNF-α明显增多(P<0.01),IL-4、IL-5明显减少(P<0.01),结肠黏膜NF-κBp50表达明显增多(P<0.01)。与模型组比较,清肠温中方各剂量组均能降低DAI评分(P<0.05),减少血清IL-1β、IL-6及TNF-α(P<0.05)、增加IL-4、IL-5(P<0.05)含量,并减少结肠黏膜NF-κBp50表达(P<0.05),尤其以清肠温中方中剂量组和美沙拉嗪组效果最明显。结论对于DSS诱导的慢性UC模型小鼠,清肠温中方可以减少Th1、增加Th2细胞,减少肠黏膜中的NF-κB表达,恢复Th1/Th2平衡从而起到治疗作用。Objective: To study the effect of Qingchang Wenzhong Decoction on balance of Th1/Th2 in chronic ulcerative colitis (UC) model mice induced by dextran sulphate sodium. Methods: 90 male BALB/c mice were divided into 6 groups randomly. Except the blank group,they drank 3% DSS solution freely for 7days,and dis- tilled water were supplied for another 7 days. One period contained 14days and 3 periods were used for copying the chronic UC model. Afterwards ,they were gavaged with corresponding medications for 14days. The disease ac- tivity index (DAI),TNF-α,IL-1β, IL-4,IL-5 and IL-6 of serum were observed by enzyme linked immunoassay and NF-KB p50 proteins were found by immunohistoehemistry. Results: Compared with the blank group, score of DAI,IL-6,IL-1β and TNF-α in serum were increased apparently (P〈0.01),while IL-4 and IL-5 were dimin- ished (P〈 0.01 ),and NF-KB in colonic mucosa was increased significantly in model group (P〈 0.01 ). Compared with the model mice, score of DAI,IL-6,IL-1β and TNF-α in serum were decreased (P〈 0.05),while IL-4 and IL-5 were increased (P〈0.05),and the expression of NF-KB was diminished in Qingchang Wenzhong Decoction groups (P〈 0.05),especially in the mild dose group and mesalazine group. Conclusion: Qingehang Wenzhong Decoction can decrease Thl ,increase Th2,and reduce NF-KB in colonic mucosa to repair the balance between them,so as to treat UC.
