摘  要:[目的]纯化T2毒素并探讨其抗肝癌活性。[方法]采用萃取法、柱层析法和高效液相色谱法从污染的玉米粒中分离T2毒素,采用MTT法、结晶紫染色法、荧光染色法考察其对两种肝癌细胞数量和形态的影响,划痕实验考察对细胞迁移的影响。[结果]纯化后,T2毒素纯度达到98%;当T2毒素浓度为8μg/mL时,对肝癌细胞的抑制率分别为(95±1.34)%(Hep G2)和(97±2.59)%(SMMC-7721),IC50分别为0.003 77μg/mL和0.003 25μg/mL,且有剂量依赖性。给药24 h后,与对照组相比,贴壁细胞减少,细胞体积缩小,肿瘤细胞迁移能力减弱。[结论]T2毒素能够成为一种潜在的抗肝癌药物。[Objective] To obtain purified T2 toxin and investigate its anti-hepatoma activity on human hepatoma carcinoma cells.[Methods] T2 toxin was extracted and purified by chromatography and HPLC.MTT staining,crystal violet staining and fluorescent staining were used to investigate the effects of T2 on proliferation and morphology of hepatoma carcinoma cells.Then the effects of T-2 on cell migration were studied using scratch experiment.[Results]Data showed that,T2 toxin displayed a significant inhibitory effects on cell proliferation and cell migration at 8 μg/mL with inhibition ratio(95±1.34) %(HepG2)and(97±2.59) %(SMMC-7721),as well as IC500.003 77 μg/mL(HepG2) and 0.003 25 μg/mL(SMMC-7721),respectively.After treated with T2 toxin for 24 h,compared with control group,hepatoma carcinoma cells showed significant shrinkage,reduction of live cells and decrease of cell migration ability.[Conclusion] T2 toxin can become a potential anti-hepatoma drug.
