摘 要:[目的]为分离鉴定山西温室蔬菜西葫芦根茎部软腐病害。[方法]对分离物进行致病性测定、形态学观察、生理生化特性测定及16SrDNA系列分析。[结果]结果表明,40个菌株经人工接种后,CY39导致西葫芦茎基部软腐,经柯赫氏法则验证为致病病原。在电镜下菌体呈短杆状,两端钝圆,具鞭毛,革兰氏阴性。CY39菌株通过测序和序列比对分析,其与胡萝卜软腐果胶杆菌胡萝卜软腐亚种(Pectobacterium carotovorum subsp.carotovorum)AB680280报告的序列相似度为100%。[结论]西葫芦茎基软腐病原菌应归属于胡萝卜软腐果胶杆菌胡萝卜软腐亚种。[Objective]Potential pathogens causing squash stem soft rot disease on greenhouse vegetables of Shanxi province were isolated and identified.[Methods]Forty isolated strains from squash were analyzed and identified by pathogenicity identification,morphological observation,determination of physiological and biochemical characteristics,and the analysis of 16 SrDNA sequence.[Results]Pathogenicity test of strains isolated from squash revealed that CY39 strain induced the disease agent.Further identification with physiological properties,16 SrDNA sequence,and PCRRFLP confirmed that isolates was P.carotovorumsubsp.carotovorum(Pcc).The 16 SrDNA testing of isolates shared99%-100% sequence similarity with that of Pcc strains.The phylogenetic trees showed that the isolates were clustered under the group of Pcc and P.chrysanthemi.and the sequence similarity between CY39 strain and AB680280 of Pectobacterium carotovorum subsp.Carotovorum was reached up to 100%.[Conclusion]To our best knowledge,this is the first report that Pcc has been proven as a causal agent of soft-rot disease on Cucurbita pepo L.
