摘  要:目的:研究连续3 d高强度间歇运动(HIIE)和中等强度持续运动(MICE)对外周血淋巴细胞凋亡的影响并探讨其可能机制。方法:16名中年久坐健康男性分别进行连续3 d HIIE(30 s全力蹬车+4 min间歇,×4组)和MICE(60%6)VO2max持续蹬车40 min),于运动前(pre)、第1 d运动后3 h(post1)、第2 d运动后3 h(post2)、第3 d运动后3 h(post3)和24 h(post24)取静脉血测定淋巴细胞计数、淋巴细胞凋亡率、淋巴细胞线粒体膜电位(MTP)、超氧阴离子、Caspase-3活性、Bcl-2蛋白表达以及血清可溶性Fas配体(s Fas L)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、C-反应蛋白(CRP)和皮质醇(CRT)含量。将新鲜淋巴细胞分别与HIIE-pre血清、HIIE-post3血清、糖皮质激素受体阻断剂米非司酮(MIF)预处理的HIIEpost3血清以及单一介质(分别为L-乳酸、hr IL-6、hr CRP和氢化可的松)在培养基中共孵育24 h,用流式细胞仪观察淋巴细胞凋亡率的变化。结果:淋巴细胞计数及比例在HIIE-post1,MTP和Bcl-2蛋白表达量在post2开始降低(P<0.05),post3达到谷值(P<0.05);淋巴细胞超氧阴离子在HIIE-post1,淋巴细胞凋亡率、Caspase-3活性和血清CRT在post2开始升高(P<0.05),post3达峰值(P<0.05);血清IL-6和CRP在HIIE和MICE后呈脉冲式升高(P<0.05),且HIIE升高幅度低于MICE(P<0.05);HIIE-post24后除Bcl-2蛋白表达仍高于pre(P<0.05)外,其他各指标均恢复(P>0.05)。HIIE-post3血清与淋巴细胞共孵育后细胞凋亡率明显高于pre血清与淋巴细胞共孵育(P<0.05)。单一血清介质与淋巴细胞共孵育显示,L-乳酸、hr IL-6和hr CRP对淋巴细胞凋亡率均无显著性影响(P>0.05),氢化可的松则上调淋巴细胞凋亡率(P<0.05),加入MIF后淋巴细胞凋亡率显著降低(P<0.05)。结论:1)连续3 d HIIE可诱导中年久坐健康男性外周血淋巴细胞凋亡,建议连续HIIE训练2 d后进行适当调整;2)线粒体氧化应激和皮质醇-糖皮质激素受体信号途径是连续HIIE诱导外周血淋Objective: This study aimed at exploring the effect of 3 consecutive days high intensity intermittent exercise( HIIE) and moderate intensity continuous exercise( MICE) on peripheral blood lymphocyte apoptosis and investigating the possible mechanism. Methods: Sixteen healthy sedentary middle-aged men performed 3 consecutive days HIIE( 4 × 30 s cycling,interspersed with 4 min rest) and MICE( 40 min continuous cycling at 60%6)VO2 max) respectively. Venous blood was taken and parameters of lymphocyte count,lymphocyte apoptosis rate,lymphocyte mitochondrial transmembrane potential( MTP),superoxide,Caspase-3 activity,Bcl-2 protein expression,and serum soluble Fas ligand( s Fas L),interleukin-6( IL-6),tumor necrosis factor-α( TNF-α),C-reaction protein( CRP) and corticosteroid( CRT) were measured before exercise( pre),3 h after the 1 st day exercise( post1),3 h after the 2 nd day exercise( post2),3 h( post3) and 24 h( post24) after the 3 rd day exercise respectively. Fresh lymphocytes and HIIE-pre serum,HIIE-post3 serum,HIIE-post3 serum preconditioning of glucocorticoid receptor blocking agent mifepristone( MIF),single mediator( L-Lactic acid,hr IL-6,hr CRP and hydrocortisone) respectively were incubated in culture medium for 24 h,and the change of lymphocyte apoptosis apoptotic rate was determined by flow cytometry. Results: Lymphocyte count and lymphocyte proportion at HIIEpost1 and MTP and Bcl-2 protein expression at post2 began to reduce( P〈0. 05),and reached the minimum level at post3( P〈0. 05). Lymphocyte superoxide at HIIE-post1,lymphocyte apoptosis rate,Caspase-3 activity and serum CRT at post2 increased and reached the peak level at post3( P〈0. 05). Serum IL-6 and CRP of HIIE and MICE increased with pulsed-wave mode( P〈0. 05),and the increase of HIIE was less than MICE( P〈0. 05). At HIIE-post24,all indicators were restored to pre-exercise( P〈0. 05),except that Bcl-2 protein was still higher( P〈
