摘 要:甲型流行性感冒是一种人群易感、症状明显、死亡率高的疾病。目前主要抗病毒药物治疗效果并不理想。热休克蛋白是机体中一类氨基酸序列高度保守,具有分子伴侣、抗凋亡、调节免疫等多种保护性功能的应激蛋白。研究发现热休克蛋白在甲型流感病毒的生命循环中起着激活病毒复制活性、稳定病毒客户蛋白等至关重要的作用。这一机制的发现为以热休克蛋白为靶点研发抗病毒药物提供了新的思路。以格尔德霉素为代表的热休克蛋白抑制剂作为重点候选药物成为了研究热点。Influenza A is a common disease with obvious symptoms and high mortality.People are vulnerable to it,but the efficacies of the current main antiviral medicines are not satisfactory.Heat shock proteins(HSPs) are stress-induced proteins with highly conservative amino acid sequences.They have many protective functions such as molecular assistance,apoptosis resistance and immune regulation.It was found that HSPs play an important role in the life circle of influenza A virus,helping activate the replication process and stabilize viral client proteins.This discovery provides a new idea for the development of antiviral drugs by focusing on HSPs.Heat shock protein inhibitors such as geldanamycin have become a research hotspot as important candidate drugs.
