摘　　要：目的：探讨2型糖尿病患者外周血TLR4、TNF-α和IL-6表达及其与疾病发生、发展的关系。方法：分别采用RT-PCR、Western blot和ELISA方法检测87例2型糖尿病患者和73例健康对照者外周血TLR4、TNF-α和IL-6表达。结果：2型糖尿病患者外周血单个核细胞TLR4的mRNA和蛋白水平表达均明显高于健康对照者，结果分别为mRNA（0．324±0．073）和（0．256±0．061）．蛋白为（0．217±0．139）和（0．165±0．094），差异有统计学意义（P〈0．05）；而血清TNF-α和IL-6表达也明显高于健康对照者，结果分别为TNF-α（2．29±0．37）和（1．25±0．12）／pg／ml，IL-6（14．28±3．9）和（9．37±2．6）／pg／ml，差异有统计学意义（P〈0．05）。结论：外周血TLR4、TNF-α和IL-6表达与2型糖尿病的发生存在一定相关性，在2型糖尿病的病程中起重要作用，为探讨2型糖尿病的发病机制和临床检测提供了新依据。Objective： To investigate the role of TLR4、 TNF-IX and IL-6 in the occurrence and development of Type 2 diabetes. Methods： Peripheral blood were isolated from 87 Type 2 diabetes patients and 73 healthy controls, the relative expression ofTLR4, TNF-α and IL-6 were detected by RT-PCR, Western blotting and ELISA. Results： The expression of TLR4 was increased in Type 2 diabetes patients. The levels ofTLR4 mRNA were （0.324±0.073） and （0.256±0.061） , and the levels of proteins were （0.217±0.139） and （0.165±0.094） , showing significant difference between the two groups （P〈0.05） . TNF-α and IL-6 were increased in Type 2 diabetes patients, the levels of TNF-α were （2.29±0.37） and （1.25±0.12） /pg/ml, and the levels of IL-6 were （14.28±3.9） and （9.37±2.6） /pg/ml, showing significant difference between the two groups （P〈0.05） . Conclusion： TLR4, TNF-α and IL-6 in peripheral blood were involved in the pathogenesis of Type 2 diabetes. This study indicated that TLR4, TNF-α and IL-6 may play an important role in Type 2 diabetes and provided a potential basis for the regulatory mechanisms in the development of Type 2 diabetes.
