健脾化痰汤对大鼠脂肪肝炎症及巨噬细胞分化的影响

摘　　要：目的:从与炎症相关的巨噬细胞分化角度探讨健脾化痰汤防治大鼠肝细胞脂肪变性的分子机制。方法:健康SD大鼠分为,正常对照组、模型组、健脾化痰组和易善复组4组,以高脂饲料联合脂肪乳灌胃复制大鼠脂肪肝模型,观察健脾化痰汤对血清C-反应蛋白、肝组织巨噬细胞(Kuppfer细胞)分化相关的炎症因子的影响。结果:健脾化痰组的肝脏指数、血清ALT、AST、C反应蛋白含量较模型组均有下降;与模型组大鼠比较,健脾化痰组血清TG含量降低、HDL/TC提高;健脾化痰组大鼠肝组织中M2型巨噬细胞的诱导因子(IL-4)及M2型巨噬细胞分泌因子(IL-10)的mRNA表达增高。结论:健脾化痰汤对高脂饮食引起的大鼠脂肪肝、血脂增高及机体炎症状态有一定的缓解作用,对肝脏的保护作用可能与促进巨噬细胞M2分化有关。Objective： Through macrophage differentiation related to inflammation to investigate the molecular mechanisms of Jianpihuatan Decoction in preventing fatty degeneration of liver cells in rats. Methods： Healthy SD rats were randomly divided into 4 groups： normal group,model group,Jianpihuatan group and Yishanfu group. The fatty-liver rat model was established by high-fat diet combined with lipid emulsion gavage. C-reactive protein of serum and inflammatory factors associated with macrophage（ Kuppfer cells） differentiation of liver tissue were observed in Jianpihuatan Decoction treated rats. Results： The liver index,levels of serum ALT,ASTand CRP in Jianpihuatan group were significantly decreased than those in model group. Jianpihuatan Decoction decreased TG and increased the ratio of HDL/TC in serum.The mRNA expressions of the inducible factors（ IL-4） and secretion factors（ IL-10） of M2 type macrophages were increased in liver tissues of Jianpihuatan group. Conclusion： The fatty liver,blood lipid increase and inflammatory state induced by high fat-diet of the rats were properly alleviated by Jianpihuatan Decoction treatment. The protective effect of Jianpihuatan Decoction on the liver might be related to promote the M2 differentiation of macrophage.
