摘  要:目的观察研究脂氧素(lipoxinA4,LXA4)抑制恶性黑色素瘤(malignant melanoma,MM)的生长及免疫机制。方法将鼠源性恶性黑色素瘤细胞B16通过皮下接种至C57BL/6小鼠腹部,以此构建MM动物模型,并通过抽签法分为作为磷酸缓冲盐溶液(PBS)组和5S,6R-7-三羟甲基庚酸(BML-111)组各6例,其中BML-111组注射LXA4类似物BML-111(1 mg/kg),PBS组则注射PBS缓冲液(1 mg/kg),两组均每两天治疗一次,共治疗5次,观察并记录肿瘤生长情况。给药结束后,称重并分别计算两组小鼠肿瘤生长抑制率。通过流式实验检测两组小鼠外周血及脾脏中淋巴细胞所占比。结果 BML-111组中肿瘤细胞生长抑制率明显高于PBS组。两组小鼠脾脏与外周血中T细胞数量比较,差异无统计学意义。结论 LXA4对MM有明显的抑制作用,但未观察到LXA4通过外周血、脾脏中T细胞免疫作用的影响。Objective To investigate the immune mechanism of lipoxinA4 and its inhibitory effect on the growth of malignant melanoma( MM).Methods The murine malignant melanoma cell line B16 was subcutaneously inoculated into the abdomen of C57 BL mice to establish an animal model of MM,and the animals were randomly divided into BML-111 and PBS groups,6 in each group.The BML-111 group was treated with BML-111( 1 mg/kg) while the PBS group was treated with PBS. The animals were treated every two day and total treatments were five times.The growth of tumor was observed and recorded. After the treatment,tumors were weighed and their growth rates were calculated to estimate the inhibitory rates of tumor growth in the both groups.The ratio of lymphocytes in peripheral blood and spleen was determined by using flow cytometry.Results The inhibitory rate of tumor growth in the BML-111 group was significantly higher than that in the PBS group.However,when calculating the relative quantities of T cell in spleen or peripheral blood,there was no significant difference between the two groups.Conclusion LXA4 inhibits the growth of MM.However,no effect of LXA4 on the T cell immunization in peripheral blood and spleen is found yet.
