摘  要:目的:通过将Melan-A抗原肽与CpG连接在DNA四面体上,并控制肽链与CpG之间的距离和CpG的个数,研究CpG对具有免疫原性的Melan-A抗原肽的协同作用。方法:将特定设计合成好的四面体-肽、四面体-肽-CpG(I)、四面体-肽-CpG(II)注射到小鼠体内,运用ELISA法检测小鼠血清中IL-6的浓度。结果:相应产生的IL-6的浓度为9.238 pg/mL、17.150 pg/mL、18.865 pg/mL。结论:CpG增强了Melan-A抗原肽的免疫作用,并且CpG个数越多增强效果越好。Objective: To study- the synergistic efteets of CpG oligodeoxynueleotides and Melan-A antigen peptide by adding a Melan-A peptide and varied numbers of CpGs to the specified positions of a DNA tetrahedron nanostrueture. Methods: Special designed DNA tetrahedron-peptide, DNA tetrahedron-peptide-CpG(I) and DNA tetrahedron-peptide-CpG (II) nanostruetures were prepared and injected into mice. ELISA method was then used to detect the level of eytokine interleukin(IL) -6 in mice serums. Results: The concentrations of IL- 6 were 9.238 pg/mL, 17.150 pg/mL, and 18.865 pg/mL, respectively. Conclusion: CpG could enhance the immunization effect of the Melan-A antigen peptide, and the effect becomes greater with increased number of CpGs added to the DNA tetrahedron nanostrueture.
