摘  要:为高效培养金线莲丛生芽,并生产大量有效物质,本研究探明了培养基中硝态氮和铵态氮浓度比对悬浮培养金线莲丛生芽有效物质积累的影响。结果表明:在MS培养基中,NO_3^-和NH_4^+浓度为30/15m mol/L时,最有利于金线莲丛生芽生物量的积累,丛生芽的鲜重和干重分别达到17.52和1.91g;有效物质的积累也最高,多糖和金线莲苷生产量分别达到7.42和1.33g/L。同时初步探究了金线莲提取物对酪氨酸酶活性的抑制效果,结果表明:金线莲丛生芽甲醇、乙醇提取物对酪氨酸酶具有较好的抑制效果,分别为54%和59%。因此,通过金线莲悬浮培养生产有效物质时,可将MS培养基中的硝态氮的浓度调节为15m mol/L,而铵态氮调节为30m mol/L,并采用金线莲丛生芽乙醇提取物可更好的抑制黑色素。In order to culture the adventitious buds of the Anoectochilus roxburghii(Wall.)Lindl,and produce the effective compounds effectively,in the study,we investigated the effects of NO3-and NH4+concentrations on effective substance accumulation,which culture in the suspension.The results showed that the accumulation of biomass of the mitiple buds is the most conducive,when the NO3-and NH4+concentrations in he MS medium of 30/15 m mol/L,which fresh weight and dry weight respectively 17.52 g and 1.91 g;at the same time,the accumulation of effective compouds achieved the highest,which production is 7.42 g/L and 1.33 g/L.This study also initially identified the inhibition of tyrosinase activity by the extract of Anoectochilus roxburghii,resulting in methanol and ethanol extracts of Anoectochilus roxburghii mitiple buds had a good inhibitory effect on tyrosinase,54% and 59%,respectively.Therefore,through used the suspension culture of Anoectochilus roxburghii to produce the effective compounds,we can theadjusted the NO3-concentration to 15 mM,while the NH4+concentration was adjusted to 30 m mol/L,and the use of ethanol extracts of clematis buds can better inhibit melanin.
