摘  要:目的研究葡萄糖酸氯己定消毒凝胶的提取方法,建立测定葡萄糖酸氯己定含量的高效液相色谱分析方法。方法研究提取溶剂、提取方法对凝胶消毒剂中葡萄糖酸氯己定回收率的影响;采用XDB-C8色谱柱,戊烷磺酸钠为离子对试剂,液相色谱法测定凝胶样品中葡萄糖酸氯己定的含量。结果采用50%乙醇水溶液为提取溶剂,60℃超声提取凝胶样品中的葡萄糖酸氯己定,葡萄糖酸氯己定在离子对反相色谱上得到基线分离,在10 mg/L^400 mg/L浓度范围为呈线性关系,仪器检出限为2.0 mg/L,日内和隔日相对标准偏差<5%,加标回收率为91.2%~99.3%。结论建立的样品提取和离子对液相色谱法线性范围宽,抗干扰能力强,适用于凝胶消毒剂中葡萄糖酸氯己定的测定,具有良好的准确度和精密度。Objective To study the extraction method of chlorhexidine gluconate and establish a high performance liquid chromatography( HPLC) method for the determination of chlorhexidine gluconate in gel disinfectant. Methods The extraction efficiency of chlorhexidine gluconate in gel disinfectant was studied by using different extraction solvent and different extraction methods. Using sodium pentane sulfonate as ion-pai reagent,XDB-C8 column was used to detect the content of chlorhexidine gluconate in gel samples. Results Chlorhexidine gluconate in the gel sample was extracted by ultrasonic extraction at 60 ℃ using 50% ethanol aqueous solution as the extraction solvent,and the content of chlorhexidine gluconat was determined by HPLC. The linear range was 10 mg/L-400 mg/L,the instrument detection limit was 2. 0 mg/L,the intraday and daytime precision was less than 5%,the recovery rate was 91. 2%-99. 3%. Conclusions The established sample extraction and reversed-phase ion-pair HPLC methods can be used to accurately determine the content of chlorhexidine gluconate in gel disinfectant. The method has the advantages of wide linear range and strong anti-interference ability,which is suitable for determination of chlorhexidine gluconat in gel disinfectant.
