摘  要:【目的】探讨青藤碱对刀豆蛋白A(Con A)诱导的大鼠急性免疫性肝损伤的防护效果及可能机制。【方法】将49只雌性Wistar大鼠随机分为7组,即正常组,模型组,青藤碱60、90、120、150 mg/kg组,地塞米松磷酸钠(Dex)组,每组7只。除正常组外,其余各组均尾静脉注射16 mg/kg Con A建立肝损伤模型。Dex组、青藤碱不同剂量组分别于Con A注射前1 h给药1次,青藤碱各组在Con A注射3 h后另再给药1次。Con A注射8 h后,腹主动脉取血,摘取肝脏组织。采用全自动生化仪检测丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、白蛋白(ALB)水平;采用酶联免疫吸附(ELISA)法检测白细胞介素2(IL-2)、白细胞介素10(IL-10)水平;采用苏木素—伊红(HE)染色观察肝组织病理改变。【结果】与正常组比较,模型组大鼠肝脾指数及ALT、AST、IL-2、IL-10水平均明显升高,ALB水平降低,差异均有统计学意义(P<0.05),表明造模成功;与模型组比较,Dex组、青藤碱不同剂量组肝指数,ALT、AST、IL-2水平均明显降低,ALB、IL-10水平升高,差异有统计学意义(P<0.05),且组织病理结果显示肝细胞坏死及炎细胞浸润程度明显减轻;与Dex组比较,青藤碱120 mg/kg组肝指数,ALT、AST、IL-2水平均明显降低,ALB、IL-10水平升高,差异有统计学意义(P<0.05)。【结论】青藤碱对Con A诱导大鼠急性免疫性肝损伤具有防治作用,抑制部分T淋巴细胞活化,调节IL-2、IL-10的分泌可能是其作用机制之一。Objective To investigate the protective effect of sinomenine(Sin)against concanavalin A(Con A)-induced acute hepatic injury in rats and to explore its possible mechanism. Methods Forty-nine female Wistar ratswere randomly divided into 7 groups, namely normal group, model group, dexamethasone sodium phosphate(Dex)group(2 mg/kg,injection through tail veins),and Sin groups(intraperitoneal injection of 60,90,120,150 mg/kg respectively),7 rats in each group. Except for the normal group,the rats in other groups were inducedinto acute liver injury model by injection with 16 mg/kg of Con A through tail veins. Dex group and Sin groups weregiven corresponding medicine one hour before Con A injection,and Sin groups were given various doses of Sinagain 3 h after injection of Con A. Eight hours after injection of Con A,blood was taken from the abdominal aortaand liver tissues were harvested. The serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and albumin(ALB)were detected by automatic biochemical analyzer. The serum levels of interleukin-2(IL-2) and interleukin-10(IL-10) were detected by enzyme-linked immunosorbent assay(ELISA), and thepathological changes of liver tissues were observed by hematoxylin-eosin staining. Results Compared with thenormal group,liver and spleen indexes,the serum levels of ALT,AST,IL-2,IL-10 in the model group weresignificantly increased,and the serum levels of ALB in the model group were significantly decreased(P〈0.05),showing the modeling was successive. Compared with the model group,liver index,the levels of ALT,AST andIL-2 in Dex group and Sin groups were significantly decreased,and the levels of ALB and IL-10 in the modelgroup were significantly increased(P〈0.05);histopathological feature results showed that the necrosis of livercells and infiltration degree of inflammatory cells were relieved. Compared with Dex group,liver index,the levelsof ALT,AST and IL-2 in Sin 120 mg/kg group were decreased,and the levels
