摘  要:【目的】探讨银翘柴桂Ⅱ号方对H1N1感染小鼠腹腔巨噬细胞Ana-1中MyD88-IRF7信号通路的影响。【方法】应用银翘柴桂Ⅱ号方对H1N1感染小鼠腹腔巨噬细胞Ana-1干预24 h后,采用酶联免疫吸附法(ELISA)测定细胞上清液中白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平,采用蛋白免疫印迹(Western blot)法检测骨髓样分化因子88(MyD88)、核因子kappa B(NF-κB)、干扰素调节因子7(IRF7)蛋白表达水平,采用反转录实时荧光定量聚合酶链反应(RT-qPCR)法检测白细胞介素1受体相关激酶4(IRAK4)m RNA表达水平。【结果】与H1N1感染组比较,银翘柴桂Ⅱ号方能显著下调Ana-1细胞IL-6、TNF-α水平(P<0.01),降低MyD88、NF-κB蛋白的表达水平(P<0.05或P<0.01),上调IRAK4 mRNA表达水平(P<0.01)。【结论】银翘柴桂Ⅱ号方可抑制H1N1所诱导的小鼠腹腔巨噬细胞My D88-IRF7信号通路以及NF-κB信号通路的活化,下调IL-6、TNF-α表达水平,发挥抗流感病毒的作用。Objective To explore the effects of Yinqiao Chaigui Recipe No.Ⅱon MyD88-IRF7 signaling pathway inmouse peritoneal macrophage Ana-1 infected by influenza A(H1N1). Methods The mouse peritoneal Ana-1 macrophages infected by H1 N1 were intervened by Yinqiao Chaigui Recipe No.Ⅱfor 24 h. And then the levels ofinterleukin(IL)-6 and tumor necrosis factor alpha(TNF-α) in cell culture supernatants were detected byenzyme-linked immunosorbent assay(ELISA), the protein expression levels of interferon regulatory factor 7(IRF7)and nuclear factor-kappa B(NF-κB)were determined by Western blotting method, and the m RNAexpression level of interleukin-1 receptor-associated kinase 4(IRAK4) was examined by real time reversetranscription quantitative polymerase chain reaction(RT-q PCR). Results Compared with H1 N1 infection group,the levels of IL-6 and TNF-α were significantly down-regulated(P〈0.01),the protein expression levels ofMy D88 and NF-κB were significantly decreased(P〈0.05 or P〈0.01),and the m RNA expression level of IRAK4 was significantly up-regulated(P〈0.01)in Yinqiao Chaigui Recipe No.Ⅱgroup. Conclusion Yinqiao Chaigui Recipe No.Ⅱ has the effects on inhibiting the activation of MyD88-IRF7 signaling pathway and NF-κB signalingpathway induced by influenza virus H1 N1,and down-regulating the expression levels of IL-6 and TNF-α,whichcontribute to the anti-virus effect.
