摘  要:目的研究坏死性小肠结肠炎(NEC)大鼠模型的简易制作方法。方法 3日龄SD大鼠30只,雌雄不限,随机分2组(每组15只),即正常对照组与NEC模型组,采用肠内配方喂养、低氧高氧暴露和冷刺激的综合应激方法造模,成功后取肠管组织,HE染色光镜下观察并行Shiou评分,Elisa法检测炎性因子(TNF-α、IL-1β和IL-6)的水平。结果 NEC模型组实验中死亡1只,对照组无死亡;与正常对照组相比,NEC模型组HE染色光镜下可见炎症反应明显,病理学检测Shiou评分明显增高(P<0.01);Elisa法检测炎性因子(TNF-α、IL-1β和IL-6)含量明显升高,差异有统计学意义(P<0.01)。结论肠内配方喂养、低氧高氧暴露和冷刺激的综合应激方法能成功建立NEC大鼠模型,且建模可重复性好,成功率高。Objective To study the simple method of making necrotic enterocolitis(NEC)rat model.Methods Thirty three-day-old SD rats were randomly divided into 2 groups(15 in each group), namelythe normal control group and the NEC model group. They were fed with enteral formula feeding, hypoxiaand hyperoxia exposure and cold stimulation. After successful stress modeling, intestinal tissue wastaken, HE-staining and light microscope was used to observe the sequential Shiou score. Elisa methodwas used to detect the levels of inflammatory factors(TNF-α, IL-1β and IL-6). Results One deathoccurred in the NEC model group and no death occurred in the control group. Compared with the normalcontrol group, the inflammatory reaction was evident under the light microscope of the HE staining in theNEC model group, and the Shiou score in the pathological examination was significantly higher(P〈0.01). The levels of inflammatory factors(TNF-α, IL-1β and IL-6)were significantly increased, andthe difference was statistically significant(P〈0.01). Conclusion The combined stress method of enteralformula feeding, hypoxia and hyperoxia exposure and cold stimulation can successfully establish the NECrat model with good repeatability and high success rate.
