摘  要:目的观察温针灸对膝骨关节炎SD大鼠行为学以及软骨中肿瘤坏死因子-α(TNF-α)和基质金属蛋白酶-3(MMP-3)的影响。方法选取SD大鼠90只,随机分为正常组、模型组和针灸组各30只,模型组和针灸组采用关节腔注射木瓜蛋白酶的方法复制右膝关节炎模型。造模成功后,针灸组给予温针灸干预2周,其余2组均不给予干预。干预前及干预2周后对3组大鼠进行Lequesne MG量表评分评估大鼠行为学表现,实验结束时处死所有大鼠,取右膝关节软骨组织,切片染色后观察,并采用放射免疫法检测软骨中TNF-α和MMP-3含量。结果干预2周后,模型组和针灸组大鼠Lequesne MG量表评分均显著低于干预前(P均<0.05),且针灸组明显低于模型组(P<0.05);针灸组膝关节病理改变明显轻于模型组,软骨中TNF-α和MMP-3含量明显低于模型组(P<0.05)。结论温针灸可以改善膝骨关节炎SD大鼠软骨形态,具有改善软骨退变和保护软骨的作用,可能与降低软骨细胞中TNF-α和MMP-3含量有关。Objective It is to observe the effects of warm-needle moxibustion on changes of behavior as well as TNF-αand MMP-3 in cartilage of SD rats with knee osteoarthritis. Methods Ninety cases of SD rats were included and were divided into three groups at random: normal group,model group and therapeutic group. In each group,there were 30 cases of SD rats. Rat knee osteoarthritis( KOA) model was established by injection of papain solution into the right knee joint. After the models were successfully established,the rats in therapeutic group were treated with warm-needle moxibustion for 2 weeks,the ones in the other two groups were given no intervention. The changes of behavior of all the rats before intervention and 2 weeks after intervention were evaluated by Lequesne MG scales. The rats were ultimately sacrificed at the end of experiment and their cartilage tissue in right knee joint was obtained and samples were sliced and dyed. Moreover,they were observed under optical microscope. Radio immunoassay method was used in measuring the level of TNF-α and MMP-3 in cartilage. Results After treatment for 2 weeks,the scores of Lequesne MG scales were significantly lower than that before treatment in model group and therapeutic group( P 〈 0. 05),the decrease of the scores in the therapeutic group was much more obvious than that in the model group( P 〈 0. 05). The pathological changes of the therapeutic group were more minimal and slighter than that of the model group. The level of TNF-α and MMP-3 in cartilage of SD rats in therapeutic group was significantly lower than that in model group( P 〈 0. 05). Conclusion Warm-needle moxibustion can help to restore the cartilage histomorphology of SD rats with knee osteoarthritis,which is also responsible for its effect in preventing and curing cartilage degeneration.
