摘  要:目的:研究水平跑台训练后生长期大鼠PBMCs中TGFβ/BMPs信号途径的Hes1、Gpnmb、Col1a1、Mmp8、Mitf、Smad7和Klf4mRNA表达水平,探索上述基因表达变化与骨重塑状态的关系。方法:24只2月龄SD雄性大鼠随机分成对照组和运动组,每组各12只。运动组以25 m/min,50 min/d,5 d/w水平运动12 w。训练结束后测量离体大鼠胫骨BMD及骨形态;用QRT-PCR检测PBMCs中Hes1、Gpnmb、Col1a1、Mmp8、Mitf、Smad7和Klf4mRNA表达。结果:与对照组相比,运动后大鼠胫骨的长度和近端BMD、BMC及厚度变化均无统计学意义。而PBMCs中Hes1、Gpnmb和Col1a1 mRNA表达上调,Mmp8 mRNA表达下调,Mitf、Smad7和Klf4表达未发生显著变化。结论:运动后PBMCs中Hes1、Gpnmb、Col1a1、Mmp8、Mitf、Smad7和Klf4 mRNA表达变化能比较敏感地反映骨重塑状态,提示上述基因可作为反应运动对骨重塑影响的候选分子标记。Objective: To explore the involvement of the TGFβ/BMPs pathway related genes Hes1,Gpnmb,Col1 a1,Mmp8,Mitf,Smad7 and Klf4 of peripheral blood mononuclear cells( PBMCs) in growing rats with treadmill training,thus to uncover the relationship between the Genes and bone remodeling caused by exercise. Methods: Two-month-old male Sprague-Dawley( SD) rats were randomly divided into two groups: control group( n = 12) and exercise group( n = 12). The rats in the exercise group received leveling treadmill running treatment 5 days per week for 12 weeks according to the exercise regimen consisted of running at 25 m/min for 50 min each day. The final training was finished. Then the bone mineral density( BMD) and the morphological change of tibia was measured. PBMCs was harvested,and the mRNA expression of Hes1,Gpnmb,Col1 a1,Mmp8,Mitf,Smad7 and Klf4 in PBMCs were detected. Results: Compared with the control group,no effect was found on the BMD and the morphological change of the tibia in exercise group. While the treadmill training up-regulated the Hes1,Gpnmb and Col1 a1 mRNA transcription,down-regulated the Mmp8 transcripts,but had no effect on Mitf,Smad7 and Klf4 transcription. Conclusion: The mRNA transcript alteration of genes Hes1,Gpnmb,Col1 a1,Mmp8,Mitf,Smad7 and Klf4 in PBMCs was more sensitive to exercise,and these genes may serve as molecular marker candidates for bone remodeling.
