摘 要:目的 观察丹参素对大鼠肝缺血再灌注损伤的保护作用.方法 60只SD大鼠随机分成3组(每组20只):假手术(Sham)组、缺血再灌注(I/R)组、丹参预处理(Sal+I/R)组.解剖出肝脏中叶、左叶的门静脉、肝动脉以及胆管,应用小血管夹钳夹后造成肝脏左、中叶缺血,1h后恢复血流,构建缺血再灌注模型.假手术(Sham)组按同样步骤解剖,但不阻断血流.丹参预处理(Sal+I/R)组于缺血前60 min经尾静脉注射丹参素40 mg/kg.再灌注6h后,各组随机各取10只大鼠,经下腔静脉收集血液标本及留取肝组织标本.再灌注12h后同样方法处理剩余大鼠.测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平.检测肝脏组织中谷胱甘肽过氧化物酶(GPX)和超氧化物歧化酶(SOD)的水平.苏木素-伊红(HE)染色,光镜观察肝脏组织病理学的变化.结果 Sal+ I/R组血清中ALT、AST、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平[6 h:(350.47 ±69.65) U/L、(500.25±76.12) U/L、(165.32±23.46) ng/L、(65.31±9.52) ng/L;12 h:(250.47±36.78) U/L、(355.25±59.12) U/L、(130.65±34.31) ng/L、(103.38±18.46) ng/L]均低于I/R组[6 h:(550.34±82.31) U/L、(702.28±106.48) U/L、(300.13±49.28) ng/L、(95.55 ±29.21) ng/L;12 h:(490.34 ±89.68) U/L、(550.28±64.23) U/L、(260.00±60.08) ng/L、(180.32±38.12) ng/L,P=0.019];Sal+ I/R组肝脏组织中GPX和SOD的水平[6 h:(52.34±11.59)、(63.00±19.23) pg/ml;12 h:(68.32±21.45)、(81.00±17.65) pg/ml]高于I/R组[6 h:(30.12±5.12)、(43.00 ±7.45) pg/ml;12 h:(39.58 ±3.16)、(54.00±8.46) pg/ml,P=0.029];光镜下Sal+ I/R组肝脏组织损伤较I/R组轻.结论 丹参素可降低大鼠肝脏缺血再灌注损伤后血清IL-6、TNF-α的水平,提高肝组织中GPX和SOD的水平,减轻缺血再灌�Objective To study the protective effect of salvianic acid A on hepatic ischemia reperfusion (I/R) injury in rats.Methods Sixty SD rats were randomly divided into 3 groups (20 rats in each group):sham operation (Sham) group,I/R group,Salvianic acid A preconditioning (Sal + I/R) group.The portal vein,hepatic artery and bile duct of the middle lobe and left lobe of the liver were dissected,and clipped by the small blood vessel clamp to make the left and middle lobe ischemia.The blood flow was restored after 1 h,and the model of I/R was constructed.In the sham operation (Sham) group,dissection was done at the same step without blocking blood flow.In Sal + I/R group,Salvianic acid A (40 mg/kg) was injected through the tail vein 60 min before ischemia.After reperfusion for 6 h,10 rats were randomly selected from each group.Blood samples were collected through the inferior vena cava and liver tissue samples were taken.After reperfusion for 12 h,the remaining rats were treated by the same method.The activities of alanine aminotransferase (ALT),aspartate amiuotransferase (AST),and interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in serum were determined.The levels of glutathione peroxidase (GPX) and superoxide dismutase (SOD) were measured in liver tissues.The pathological changes of liver tissue were observed by hematoxylin and eosin (HE) staining and light microscopy.Results The levels of ALT,AST,IL-6 and TNF-α in the Sal + I/R group [6 h:(350.47 ± 69.65),(500.25 ± 76.12) U/L,(165.32±23.46),(65.31 ±9.52) ng/L;12 h:(250.47 ±36.78),(355.25 ±59.12) U/L,(130.65 ±34.31),(103.38 ± 18.46) ng/L were lower than those in the group I/R [6 h:(550.34 ±82.31),(702.28± 106.48) U/L,(300.13 ±49.28),(95.55 ±29.21) ng/L;12 h:(490.34±89.68),(550.28 ±64.23) U/L,(260.00 ±60.08),(180.32 ±38.12) ng/L,P =0.019];The levels of GPXand SOD in the Sal +L/R group [6 h:(52.34± 11.59),�
