摘  要:目的观察生麦芽对利培酮所致高泌乳素血症(HPRL)雌性大鼠血清泌乳素(PRL)水平和垂体PRL mRNA表达的影响。方法大鼠灌胃利培酮口服液每日0.1 mg·kg^(-1)、连续30 d制备HPRL模型。造模后,按照体重将造模成功大鼠随机分为4组,每组10只:模型组和低、中、高3个剂量实验组。4组大鼠每日灌胃利培酮0.1 mg·kg^(-1);另外,低、中、高3个剂量实验组每日分别灌胃生麦芽汤剂10,20,40 g·kg^(-1)。正常组每日灌胃相同体积的0.9%Na Cl,均连续45 d。酶联免疫吸附测定法测定大鼠血清PRL水平;用逆转录聚合酶链式反应测定大鼠脑垂体组织中PRL mRNA表达水平,计算各组大鼠垂体PRLmRNA表达的倍数。结果正常组、模型组和低、中、高3个剂量实验组大鼠血清PRL的含量分别为(16.75±2.55),(17.14±1.81),(14.15±3.02),(13.65±2.28),(14.45±1.71)pg·m L^(-1),低、中、高3个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。以正常组垂体PRLmRNA的表达倍数为1,模型组和低、中、高3个剂量实验组的表达倍数分别为1.26±1.21,4.29±1.78,5.42±0.76,4.50±0.29倍。低、中、高3个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论利培酮可以引起雌性大鼠血清PRL升高;生麦芽可以降低由利培酮所致雌性大鼠的HPRL。Objective To study the effect of raw malt on contents of prolactin( PRL) in the serum and the of expression of PRL mRNA in hypophysis of female rats hyperprolactin( HPRL) induced by risperidone. Methods The rats were fed with risperidone 0. 1 mg·kg^-1·d^-1.After 30 d,the HPRL model was prepared. Then the rats were randomly divided into four groups according to their weight,each group had 10 rats: model group and experimental-L,experimental-M,experimental-H groups. And continued to be filled with risperidone 0. 1 mg·kg^-1·d^-1 in 4 groups. In addition,the experimental-L,experimental-M,experimental-H groups were also fed with raw malt 10,20,40 g · kg^-1· d^-1. Equal volume of 0. 9% NaCl was given to normal group. After 45 d,serum PRL levels in rats were measured by enzyme linked immunosorbent assay. Reverse transcription polymerase chain reaction( RT-PCR) was used to measure hypophysis PRL mRNA expression,and calculate the difference multiples of expression. ResultsSerum PRL levels were( 16. 75 ± 2. 55),( 17. 14 ± 1. 81),( 14. 15 ± 3. 02),( 13. 65 ± 2. 28),( 14. 45 ± 1. 71)pg·m L^-1 in normal group,model group,experimental-L,experimental-M,experimental-H group; comparison between drugs groups and model group,the difference had significantly( all P〈0. 05). The hypophysis PRL mRNA expression was 1,then 1. 26 ± 1. 21,4. 29 ± 1. 78,5. 42 ± 0. 76,4. 50 ± 0. 29 in normal group,model group,experimental-L,experimental-M,experimental-H group; comparison between three experimental groups and model group,the difference had significantly( all P〈0. 05). Conclusion The serum PRL level in female rats can be increased by risperidone. The raw malt can treat the HPRL of female rats caused by risperidone.
