摘  要:以奶牛乳腺上皮细胞为材料,在克隆PRSS23基因的基础上,构建真核过表达栽体和干扰栽体并将其转染到奶牛乳腺上皮细胞中,利用qRTPCR的方法检测PRSS23基因在mRNA水平上的表达情况,同时测定细胞内甘油三酯的含量。结果显示:在mRNA水平上,过表达组与对照组相比表达量差异显著,其甘油三酯水平显著升高;干扰组与对照组相比,在mRNA水平上差异表达显著,甘油三酯含量也显著升高。On the basis of cloning PRSS23 gene and using mammary gland epithelial cells as mate- rials,we transfected eukaryotic overexpression vector and interference vector which we constructed into mammary epithelial cells of dairy cow. The expression of PRSS23 gene at the mRNA level was detected by qRT-PCR and the intracellular triglyceride content was measured in cells. The results showed that the expression levels in the overexpression group compared with the control group were significantly different at the mRNA level, and the triglyceride level was significantly increased. The difference at mRNA level between the interference group and the control group was significant,and the triglyceride content was also significantly increased.
