摘 要:将40只泌乳期母鼠随机分为5组(n=8)。除空白对照组(I组)不做任何处理外,其余的在产后3~5d经乳导管分别向第4对乳头内注射20μL无菌生理盐水(阴性对照组,Ⅱ组)和低(Ⅲ组,1×10 7CFU/mL)、中(Ⅳ组,1×10 8cFu/mL)、高(V组,1×10 9CFU/mL)浓度大肠杆菌和金黄色葡萄球菌混合菌悬液。并于0,24,48,72h观察各组小鼠临床症状、组织病理学变化、乳腺的间质宽度及腺泡壁厚度和乳腺组织匀浆中TNF-α等炎症相关因子及菌落CFU含量变化。结果显示:与空白对照组相比,模型组随着注射细菌浓度增大而呈现乳腺组织病变加重;乳腺间质增宽及腺泡壁厚度减小、乳腺组织匀浆中的TNF-α、IL-6及CFU含量显著增高且差异显著。并以1×10 8CFU/mL组炎症变化最完整,病理变化适中、不出现死亡,且小鼠病理模型的炎症病理过程和炎症反应相一致,可以诱发典型小鼠乳腺炎病理模型。结果表明:1×10 8CFU/mL组可选为模型组。Forty lactating female mice were randomly divided into five groups(n = 8). Except the blank control group(group Ⅰ )without any treatment,in the remaining four groups 20 μL of sterile physiological saline(group Ⅱ )or different concentration of Escherichia coli and Staphylococcus aureus mixed bacteria suspension(group Ⅲ 1 × 10 7 CFU/mL,group Ⅳ 1× 10 8 CFU/mL,group V 1 × 109 CFU/mL)were injected into the fourth abdominal mammary gland via the teat duct at 3-5 d after parturition respectively. The clinical-symptoms, histopathological evaluation, interstitial width,alveoli cases thickness and colony for units(CFU), tumor necrosis factor-α(TNF-α) concentration in the mammary glands tissue homogenate were observed at 0,24,48 and 72 h. The results show that compared with the blank control group, the model groups increased breast lesions with the increased injection of bacterial concentrations increased interval width and decrease alveoli ca- ses thickness. The contents of TNF-α, IL-6 and CFU in the mammary glands tissue homogenate were significantly higher in group Ⅲ , Ⅳ and Ⅴ than those in group Ⅳ (P〈0.05). In addition,the pathological changes of group IV were moderate and no death occurred. The pathologic model of mice was consistent with inflammatory pathological process and inflammatory reaction, indicating inducing a pathological model of typical mastitis in mice.
