电针肝郁脾虚功能性消化不良大鼠血清代谢组学分析

摘　　要：目的：从代谢组学的角度,探讨肝郁脾虚型功能性消化不良（FD）病理机制,以及电针治疗的作用机理。方法：选用21只大鼠分为空白组、模型组和电针组。实验前14 d进行肝郁脾虚FD大鼠的造模;造模成功后,从第15-28天,对电针组给予电针针刺足三里和肝俞穴的干预,观察大鼠行为学变化,计算胃内残留物率和小肠推进率检测胃肠道运动功能;采用高效液相色谱-质谱联用（LC-MS）方法对各组血清进行代谢组学分析,比较组间差异及潜在标志物。结果：电针组胃肠道运动功能明显改善,空白组、模型组和电针组大鼠血清中VIP〉1的物质被筛选出来,通过两两比较分析代谢物差异（P〈0.05）,共有11种物质为潜在的生物标记物。结论：电针相应穴位可能通过影响脑肠轴、炎症反应、免疫反应等多种途径共同作用,来修复代谢紊乱,从而改善肝郁脾虚型大鼠FD症状。Objective： To explore the pathological mechanism of liver depression and spleen deficiency syndrome functional dyspepsia,and the mechanism of electroacupuncture therapy by metabonomics. Methods： Twenty-one rats were divided into blank group,model group and electroacupuncture group. Modeling of FD rats with liver qi stagnation and spleen deficiency syndrome was set up 14 d before experiment. After the success of the model making,from 15 d to 28 d,electroacupuncture group was given to the electroacupuncture at Zusanli（ST36） and Ganshu（BL18）. The behavior changes of rats were observed and the gastrointestinal motility was measured by calculating the rate of gastric remnants and the propulsive rate of the intestine. We compared the difference serum metabolites in each group,and analyzed the potential markers by LC-MS method. Results： The gastrointestinal motility was obviously improved in the electroacupuncture group. The VIP〉 1 substances in the serum of the rats in the blank group,the model group and the electroacupuncture group were screened out. The difference of metabolites was analyzed by pair comparisons（P〈0. 05）,and 11 substances were found as potential biomarkers. Conclusion： Electroacupuncture at corresponding points may affect brain-gut axis,inflammatory response and immune response. Many pathways are combined to repair metabolic disorders and improve FD symptoms in rats with liver depression and spleen deficiency syndrome.
