摘  要:水稻白叶枯病是最重要的水稻细菌性病害,严重威胁水稻生产和粮食安全。培育和种植抗白叶枯病水稻新品种是控制该病害最经济、有效、环保的技术手段。W6023是本课题组早期通过普通野生稻与IR24杂交创制的抗病基因导入系,前期转录组测序结果显示,Pong2-1(Os02g20780)和Pong11-1(Os11g14160)在感病亲本IR24中激活表达,在抗病导入系W6023中不表达。Pong2-1和Pong11-1是水稻中Pong类转座子最相似的两成员,DNA同源性超过90%,但转录本却存在明显差异。为研究Pong2-1和Pong11-1是否与IR24的感病性相关,本研究利用CRISPR/Cas9系统定点突变IR24的Pong2-1和Pong11-1位点,获得了多个突变株系,毒力测试结果显示Pong2-1和Pong11-1突变后IR24的抗性得到一定提高,创制出6个抗白叶枯病水稻材料。Bacterial blight of rice, caused by Xanthomonas oryzae pv. oryzae ( Xoo), is one of the most destruc- tive diseases of flee, which seriously affects the rice production in Asian countries. Deployment of resistance genes in varieties is the most effective, economic and environment-friendly strategy for disease control. Previously, we generated a Xoo-resistant introgression line W6023 that derived from a cross between a wild rice accession and susceptible eultivar IR24. The transcriptome analysis detected the expression of two genes Pong2-1 (OsO2g20780) and Pongll- 1 (Osl 1g14160) in susceptible line IR24 ,but not in resistant line W6023. Despite of 〉 90% sequence similarity, the genes Pong2-1 and Pongll-1 represented different forms of transcripts. To investigate whether these two genes are susceptibility factors to Xoo, we knocked out Pong2-1 and Pong11-1 in susceptible line IR24 using CRISPR/ Cas9 methods and obtained a series of mutant lines. Test for resistance to PXO99A showed enhanced resistance in the double mutant lines ,which served as resistant resource potentially being applied in rice breeding.
