中药复方多糖对不同MHC B-LβⅡ基因型鸡淋巴细胞IL-1β、IL-8和TNF-α mRNA表达量的影响

摘　　要：探讨中药复方多糖对不同MHC B-LβⅡ基因型鸡淋巴细胞IL-1β、TNF-α、IL-8mRNA表达量的影响。采用PCR-SSCP方法将500羽京红1号蛋鸡按不同MHC B-LβⅡ基因型分组,采集不同MHC BLβⅡ基因型鸡的外周血淋巴细胞,分别加入终浓度为100、75、50、0μg/mL中药复方多糖共培养24h,收集细胞,采用real-time PCR方法检测不同MHC B-LβⅡ基因型鸡IL-1β、TNF-α、IL-8基因mRNA的表达。结果显示,与对照组相比,不同剂量中药复方多糖均能提高不同MHC B-LβⅡ基因型鸡IL-1β、TNF-α、IL-8基因mRNA的表达量(P<0.05)。中药复方多糖剂量为50μg/mL时,AA、BC基因型鸡IL-1β、TNF-α、IL-8基因mRNA的表达量最高,中药复方多糖剂量为100μg/mL时,BB基因型鸡IL-1β、TNF-α、IL-8基因mRNA的表达量最高。中药复方多糖能不同程度的促进各MHC B-LβⅡ基因型鸡IL-1β、TNF-α、IL-8基因mRNA的表达,且不同MHC B-LβⅡ基因型鸡淋巴细胞最适中药复方多糖免疫调节剂量略有不同。To investigate the effects of compound Chinese herbal medicinal polysaccharides on the expressions of IL-1β,IL-8,TNF-α mRNA in lymphocytes of chickens with different MHC B LβII genotypes,PCR-SSCP method was used to group 500 layers of Jinghong 1 according to different MHC B-LβII genotypes.The peripheral blood lymphocytes of different MHC B-LβII chickens were collected and the compound Chinese herbal medicinal polysaccharides were added to the final concentrations of 100,75,50,0 g/mL respectively,and cultured for 24 hours.The cells were collected.The expressions of IL-1β,TNF-α,IL-8 mRNA in lymphocytes of chickens with different MHC B-LβII genotypes were detected by real-time PCR.The results showed that compound Chinese herbal medicinal polysaccharides at different does could improve mRNA expressions of IL-1β,TNF-α,IL-8 genes in chickens with different MHC B-LβII genotypes（P〈0.05）;When the dosage of compound Chinese herbal medicinal polysaccharides was 50μg/mL,the expressions of mRNA of IL-1β,TNF-α,IL-8 genes were the highest in the AA BC genotype chickens,and the highest expressions of mRNA levels were found in the IL-1β,TNF-α,IL-8 genes in the chickens when the dosage of compound Chinese herbal medicinal polysaccharides was 100μg/mL.Compound Chinese herbal medicinal polysaccharides can promote the expressions of mRNA of IL-1β,TNF-α,IL-8 genes in different MHC B-LβII genotypes of chickens,and the most suitable dosage of compound Chinese herbal medicinal polysaccharides for immune regulation of lymphocytes in different MHC B-LβII genotype chickens is slightly different.
