天花粉多糖的组成及组分Ⅱ相对分子质量分析

摘　　要：目的:对天花粉多糖的组分及组分Ⅱ相对分子质量进行分析。方法:采用水提醇沉提取天花粉多糖,经纯化后进行气相-质谱联用分析得出多糖的组成。相对分子质量分析则采用高效凝胶渗透色谱法(HPGPC)。结果:天花粉多糖组分Ⅰ是含有葡萄糖,未知单糖一和未知单糖二的杂多糖。天花粉多糖组分Ⅱ是含有鼠李糖、甘露糖、葡萄糖、半乳糖和未知单糖三的杂多糖。天花粉多糖组分Ⅱ经高效凝胶渗透色谱法得重均相对分子质量(Mw)=42 025;10%大分子部分重均相对分子质量=197 011;10%小分子部分重均相对分子质量=6 676;分布系数(D)=2.56;数均相对分子质量(Mn)=16 427。结论:天花粉多糖组分Ⅰ和天花粉多糖组分Ⅱ的单糖组成不相同,天花粉多糖组分Ⅱ是一个相对分子质量段在42 025左右的大分子。OBJECTIVE To analyze the polysaccharide composition and molecular weight of the polysaccharide componentⅡof trichosanthis radix.METHODS The polysaccharide of trichosanthis radix was extracted by water extraction and alcohol precipitation.After purification,the composition of polysaccharides was analyzed by gas chromatography and mass spectrometry.High performance gel permeation chromatography（HPGPC）was used for molecular weight analysis.RESULTS Polysaccharide componentⅠcontained glucose,unknown monosaccharide one and unknown monosaccharide two.Polysaccharide componentⅡcontained rhamnose,mannose,glucose,galactose and an unknown monosaccharide.By gel permeation chromatography method,the weight-average molecular weight（Mw）of polysaccharide componentⅡ was 42 025;10% molecular weightaverage molecular weight was 197 011;small molecular weight-average molecular weight was 6 676;distribution coefficient（D）was 2.56;number-average molecular weight（Mn）was 16427.CONCLUSION The compositions of polysaccharide componentⅠ and polysaccharide componentⅡ of trichosanthis radix are different in monosaccharide composition.Polysaccharide componentⅡ was a large molecule with a molecular weight of about 42 025.
