摘  要:目的:探讨细颗粒物(PM2.5)对慢性阻塞性肺疾病(COPD)模型大鼠血清及肺泡灌洗液(BALF)中髓过氧化物酶(MPO)表达的影响。方法:SPF级2月龄雄性Wistar大鼠32只按照随机数字表法分为对照组、PM2.5组、COPD组、COPD+PM2.5组,每组各8只。采用香烟烟雾暴露法建立COPD模型,在造模成功后第1、3、5天,PM2.5组及COPD+PM2.5组气管滴注PM2.5混悬液共3次(10 mg/kg),在最后一次气管滴注完成24 h后处死各组大鼠,留取肺组织标本行病理学检测,采用ELISA方法检测血清及BALF中MPO的表达。结果:与对照组相比,COPD组气道壁及平滑肌明显增厚,肺泡腔扩大、肺泡壁变薄、部分破裂融合成肺大疱,肺泡数目显著减少;PM2.5组气道壁及平滑肌无明显增厚,但有肺泡壁破坏;COPD+PM2.5组气道壁及平滑肌增厚、肺泡壁破坏更明显。COPD组、PM2.5组和COPD+PM2.5组血清及BALF中MPO表达均较对照组增加(P<0.01),且COPD+PM2.5组血清及BALF中MPO表达水平均高于COPD组与PM2.5组(P<0.05)。结论:PM2.5加剧了COPD模型大鼠肺泡壁的破坏,这可能与中性粒细胞参与的炎症反应有关。Objective:To investigate the effects of fine particulate matter(PM2.5)on the expression of myeloperoxidase(MPO)in serum and bronchoalveolar lavage fluid(BALF)in chronic obstructive pulmonary disease(COPD)rats. Methods:A total of 32 male rats were randomly divided into control group,PM2.5 group,COPD group,and COPD + PM2.5 group. COPD model was established by cigarette smoke exposure. At 1,3 and 5 days after COPD model established,PM2.5(10 mg/kg)was intratracheally instilled in PM2.5 and COPD+PM2.5 groups for three times. The rats were sacrificed at 24 h after the last intratracheal instillation and lung tissue was kept to do pathological examination. Expression levels of MPO in serum and BALF were measured by ELISA method. Results:Compared with control group,the pathology of COPD group showed obvious incrassation of airway wall and smooth muscle,enlargement of alveolar space,thinning of alveolar wall,formation of emphysema,and significant reduction of the number of alveoli.The PM2.5 group showed no obvious incrassation of airway wall and smooth muscle,but damage of some alveolar wall. The COPD+PM2.5 group showed more obvious incrassation of airway wall and smooth muscle,and more damage of some alveolar wall. Expression levels of MPO in serum and BALF in COPD group,PM2.5 group and COPD+PM2.5 group were obviously higher than those in control group(P〈0.01). Expression levels of MPO in serum and BALF in COPD+PM2.5 group were obviously higher than those in COPD group and PM2.5 group(P〈0.05). Conclusion:PM2.5 can damage alveolar wall in COPD rats,and this is closely associated with the macrophages involved inflammatory reaction.
