摘 要:目的:探讨甲基苯丙胺(MA)对大鼠海马组织骨架肌动蛋白表达的影响,为临床研究提供数据支持。方法:选取健康雄性Wistar大鼠60只,随机分为实验组和对照组,实验组分别给予不同剂量的MA制作1、2、4周依赖模型,每组10只;对照组按照相同剂量给予生理盐水。判定依赖模型制作成功后,于最后一次给药5 h后将大鼠断头处死,常规分离海马组织,采用BradFord法检测大鼠海马组织肌动蛋白和结合蛋白的表达水平。结果:MA依赖1、2、4周组的G-actin表达量与相应对照组比较差异均无统计学意义(P>0.05);MA依赖1、2周组的F-actin表达量与相应对照组比较差异均无统计学意义(P>0.05),依赖4周组的F-actin表达量显著高于相应对照组(P<0.05);MA依赖1周组的p-cofilin表达量与相应对照组比较差异无统计学意义(P>0.05),2周组和4周组的p-cofilin表达量均显著高于相应对照组(P<0.05)。结论:MA对大鼠海马神经细胞骨架肌动蛋白的表达具有直接的影响。Objective:To investigate the effects of methamphetamine(MA)on the expression of the cytoskeleton actin in rat hippocampal tissue and provide data support for clinical research. Methods:A total of 60 Wistar male rats were randomly divided into the experimental group and the control group. In the experimental group,1 week,2 weeks and 4 weeks MA-dependent models were established with different dose of MA(10 rats per group). In the control group,rats were given with the equal volume of saline. MAdependent models were established successfully,then the rats were executed and the hippocampus were separated at 5 h after the last administration,and the expression levels of the actin and actin binding proteins were detected with BradFord. Results:There was no significant difference in G-actin expression of 1 week,2 weeks and 4 weeks MA-dependent groups compared with corresponding control group(P 〉 0.05). There was no significant difference in F-actin expression of 1 week,2 weeks MA-dependent groups compared with corresponding control group(P 〉 0.05). The F- actin expression of 4 weeks MA- dependent group significantly increased compared with control group(P 〈 0.05). There was no significant difference in p-cofilin expression of 1 week MAdependent group compared with corresponding control group(P 〉 0.05),while the p-cofilin expression of 2 weeks and 4 weeks MAdependent groups significantly increased compared with control group(P 〈 0.05). Conclusion:There is direct effect of MA on the cytoskeleton actin rearrangement in rat hippocampal nerve cells,which provides reference value for clinical application and research of medicine.
