摘  要:组培技术对于近几年新兴的蓝莓产业来说具有繁殖系数高、不受季节限制、短期内可获得大量繁殖体等优点,该方法是目前解决蓝莓产业化发展中种苗短缺的关键技术,但是蓝莓生根难的问题制约了组培技术的广泛应用。为进一步探究生长素种类和浓度对蓝莓生根的影响,本研究以奥尼尔、乔治、都克及美登这4个蓝莓品种当年新生嫩枝作为外植体建立了无菌株系,在此基础上经过扩繁5~7代并进行壮苗培养。以1/2WPM为基本培养基,通过添加不同种类和浓度的生长素(IBA和NAA),选用生根率、生根数及根长等指标评价生长素对不同基因型蓝莓生根的影响。研究结果表明,对不同基因型蓝莓来说,其生根难易程度不同。对于同一品种的蓝莓,其生根率、生根数及根长随着IBA浓度的不同而有所变化。综合分析认为蓝莓4个品种的最佳生根培养基配方为1/2 WPM+2.0 mg/L IBA+0.2%活性炭。本研究为优化蓝莓组培技术,筛选出适宜北京地区生长的蓝莓,以及为北京地区蓝莓的产业化发展提供了技术支持。The tissue culture technique has advantages of high reproduction coefficient,no seasonal restriction,and fast cultivation in short-term for blueberry industry which has been flourishing in recent years.In addition,the techniques has become the key technology to solve the shortage of seedlings in blueberry industrialization.However,the difficulty in blueberry rooting restricts the wide application of tissue culture.In order to further investigate the effects of different auxins and its concentrations on rooting of blueberry,the research in this paper had built sterile strains of O'Neal,George,Duke and Blomidon,and cultivated the strains for 5 to 7 generations to make it stronger.Then,this experiment used 1/2 woody plant medium(WPM) as basic medium and the variation of roots was controlled by different varieties and concentrations of auxin(IBA and NAA).We used rooting rate,root number and root length as indicators to evaluate the effects of rooting for different genotypes of blueberry.The results showed that the different genotypes of blueberry had different difficulties in rooting.For the same variety of blueberries,the rooting rate,root number and root length varied with the concentrations of IBA.The comprehensive analysis suggested that the best rooting medium for these 4 varies was 1/2 WPM +2.0 mg/L IBA +0.2% acticarbon.This study could provide technical supports for optimization of blueberry tissue culture techniques,screen of the best growth region in Beijing,as well as the industrialization development of blueberry in Beijing.
