摘 要:为研究优质肉鸡M系NPY、NCOA-1、IGF-1和GDF-9基因聚合与繁殖性能的关联性分子标记,进一步提升多基因聚合技术在优质肉鸡分子育种中的应用普及率,试验采用SNP分型检测和x2检验等生物统计方法分析了NPY、NCOA-1、IGF-1和GDF-9基因多态性及优质肉鸡多基因聚合后混合基因型与繁殖性能的关联性。结果表明,优质肉鸡M系4个基因均有多种基因型,其中NPY、IGF-1和GDF-9基因SNP位点所在受试群体达到Hardy-Weinberg平衡,NCOA-1基因所在受试群体未达到Hardy-Weinberg平衡。Aa BBCCGg基因型在开产日龄上显著低于其他基因型(P<0.05),在开产体重上显著高于AABBCCGg(P<0.05),在300日龄合格种蛋率上显著高于其他基因型(P<0.05);AABBCCGg在开产蛋重、300日龄蛋重上显著低于其他基因型(P<0.05),而aa BBCCGg在300日龄产蛋量上优势显著(P<0.05)。聚合基因型Aa BBCCGg基因型在开产日龄、开产体重和300日龄合格种蛋率上显著优于其他基因型(P<0.05),AABBCCGg在开产蛋重、300日龄蛋重上显著低于其他基因型(P<0.05),而aa BBCCGg在300日龄产蛋量上优势显著(P<0.05)。综合比较得出Aa BBCCGg聚合基因型为最有利聚合基因型,可作为有效改良优质肉鸡繁殖性能的候选标记。This experiment was conducted to evaluate the molecular markers associated with the relation between the reproductive performance and the NPY, NCOA-1, IGF-1, and GDF-9 genes using the multiple gene pyramiding technology in the high quality broiler M line and to further enhance the application of the multiple gene pyramiding technology in high quality broiler molecular breeding. SNP technique and x2 examine were used to analyze the polymorphism and relation between the reproductive performance and the multiple gene pyramiding of NPY, NCOA-1, IGF-1, and GDF-9. The results showed that: four genes in the high quality broiler M line had a variety of genotypes, of which, NPY, IGF-1, and GDF-9 genotypes met the Hardy-Weinberg equilibrium, while NCOA-1 genotype did not reach the Hardy-Weinberg equilibrium. The polymeric genotype Aa BBCCGg was significantly lower in first laying age than others(P〈0.05), but higher than AABBCCGg in first laying body weight(P〈0.05) and higher than the other genotypes in settable eggs of 300 day-old(P〈0.05). The polymeric genotype AABBCCGg in the egg weight and laying rate of the 300 day-old were significantly lower than the others(P〈0.05). The polymeric genotype aa BBCCGg was higher than others in the laying rate of 300 day-old(P〈0.05). Compared with all genotypes, we concluded that the polymeric genotype Aa BBCCGg was the best one, which can be used as a candidate marker to effectively improve the breeding performance of quality broilers.
