摘  要:目的验证常规Hi-C(highest-throughput chromosome conformation capture)建立文库的方法,并留意其中的关键步骤和注意事项,为以后的研究者提供参考依据。方法收集染色体核型异常者和正常者各2例的外周血标本,提取单个核细胞,经甲醛交联后,按照常规Hi.c方法,构建Hi-C文库,并对实验过程进行浓度检测、交联效果检测、染色质完整性检测、文库质量检测等质量控制。结果构建了4个Hi-C文库,对常规Hi-C流程的各个质量质控点的检测结果均符合Hi-C建库的要求。结论常规Hi-C流程的可重复性是可靠的。Objective To verify the method of establishing a library for highest-throughput chromosome conformation capture (Hi-C) and pay attention to the key steps and precautions there- in, and provide reference for future researchers. Methods Peripheral blood samples were collected from two patients with abnormal karyotype and two cases with normal karyotype, and mononuclear cells were extracted. After cross-linked by formaldehyde, Hi-C library was constructed according to the conventional Hi-C method. After cross-linking, Hi-C library was constructed, cross-linking effect and chromatin integrity were detected; library quality control and other quality control were performed. Results Four Hi-C libraries were constructed. The test results of each quality control point of the routine Hi-C flow met the requirements of Hi-C database construction. Conclusion The repeatability of a conventional Hi-C flow is reliable.
