摘 要:目的:观察芩百清肺浓缩丸(芩百)对感染后咳嗽(PIC)大鼠肺泡灌洗液和血清SP、CGRP及NEP含量及肺组织SP及NEP表达的影响,探讨芩百清肺浓缩丸对感染后咳嗽气道神经源性炎症的干预作用。方法:40只大鼠随机分为正常组、模型组、阳性对照组(孟鲁司特)和中药芩百组。采用烟熏加内毒素(LPS)滴鼻以及辣椒素雾化吸入法建立PIC大鼠模型。各组大鼠给予相应处理10 d,观察大鼠3 min咳嗽次数变化,ELISA法检测BALF、血清中SP、CGRP和NEP含量,免疫组化检测肺组织SP、NEP蛋白表达并进行半定量评分,HE染色光镜观察支气管和肺组织病理改变。结果:1)咳嗽次数比较:阳性对照组和中药组咳嗽次数低于模型组,差异有统计学意义(P<0.05),两药物组间比较,差异无统计学意义(P>0.05)。2)SP、CGRP及NEP含量比较:中药组BALF、血清中SP和CGRP含量均低于模型组和阳性对照组,差异有统计学意义(P<0.05);中药组BALF中NEP含量接近正常组水平,高于模型组和阳性对照组,差异有统计学意义(P<0.05);各组血清中NEP含量比较,差异无统计学意义(P>0.05)。3)肺组织SP与NEP免疫组化检测结果显示:与模型组比较,中药组SP表达明显减弱、NEP表达增强,差异有统计学意义(P<0.05);阳性对照组与模型组比较,差异无统计学意义(P>0.05)。结论:芩百清肺浓缩丸能够降低感染后咳嗽大鼠血清及BALF中SP、CGRP的含量及肺组织SP的表达,提升BALF中NEP的含量及表达至接近正常水平。芩百清肺浓缩丸能够且不仅仅是通过NEP-SP途径来降低感染后咳嗽的气道神经源性炎症。Objective: To observe the effects of Qinbai Qingfei Pill(Qinbai) on the content of SP,CGRP and NEP of BALF and serum,the protein expression of SP and NEP in lung tissue of rats with PIC,investigated the role of Qinbai on airway neurogenic inflammation in PIC. Methods: 40 rats were divided into normal,model,positive control and Qinbai groups. After establishing the PIC model by smoke add intranasal with LPS and atomization inhalation with capsaicin,each group was given the corresponding medicine or saline for 10 d. The following indexes were observed,the number of cough in 3 min,the content of SP、CGRP and NEP in serum and BALF by ELISA,the protein expression of SP and NEP in lung tissue by immunohistochemistry,the pathology changes of lung tissue also be observed by light microscopy. Results: 1) The number of cough: It was fewer in the positive control group and Qinbai group that that of the model group,the differences were statistically significant(P〈 0. 05). The difference between the positive control group and Qinbai group was not statistically significant(P 〉0. 05). 2) SP、CGRP and NEP contents in serum and BALF : The contents of SP and CGRP of serum and BALF in the Qinbai group were lower than those of the positive control and the model groups,the differences were statistically significant(P〈 0. 05);the content of NEP of BALF in the Qinbai group was closed to normallevel,that was higher than that in positive and model groups,the differences were statistically significant(P〈 0. 05);but the differences of serum NEP content in eachgroup were not statistically significant(P〉 0. 05). 3) SP and NEP expressions in lung tissue: The expression of SP was weakened substantially,while the expression of NEP was strengthened in the Qinbai group comparing with the model group,the differences were statistically significant(P 〈0. 05),the differences of these expressions between the positive control group and the model group were not statistically significant(P 〉0. 05).
