摘  要:目的:探讨酪蛋白激酶2相互作用蛋白1(Ckip-1)对小鼠颅颌面耳廓软骨发育的影响。方法:取6月龄Ckip-1基因敲除小鼠(Ckip-1-/-)作为实验组,野生型小鼠(WT)作为对照组(n=6、同窝、雄性)行动物表型观察及组织学切片分析,评估Ckip-1-/-小鼠耳廓大体及组织学变化。另通过siRNA干扰技术下调小鼠软骨细胞系ATDC5中Ckip-1表达,通过扫描电镜、MTT及QPCR等实验,观察细胞形态、增殖能力及软骨细胞分化相关基因表达的变化。结果:Ckip-1-/-小鼠呈小耳畸形,组织学切片可见软骨结构紊乱,幼稚软骨细胞增多。Ckip-1敲低组软骨细胞体现出伸展面积增加、增殖能力下降、软骨早期分化因子增加及晚期分化因子下调等趋势。结论:Ckip-1基因可能通过调控软骨细胞增殖及分化对小鼠耳廓发育产生影响。Objective: To investigate the effects of Casein kinase 2 interacting protein-1 (Ckip-1) on the development of auricular cartilage in mice. Methods: 6-month-old mice with Ckip-1 knockout( Ckip-1^-/- ) were used as the experimental group, wild type mice(WT) as the control group(n = 6, same litter and gender, male). The anatomical and histological feature of auricular cartilage was studied by gross observation and histological examination. Ckip-1 was down-regulated in ATDC5 chondrocytes by siRNA interference technique. Scanning electron microscope, MTT and QPCR assays were used to investigate the cell morphology, proliferation, and chondrocyte differentiation related gene expression. Results: Ckip-1^-/- mice showed auricular cartilage deformities and structure disorder, more immature chondrocytes in the cartilge. Down-regulation of Ckip-1 in ATDC5 chondrocytes increased the extensional area of the cells and the expression of early ehondrocyte differentiation factors, the cell proliferation ability and the late differentiation factors were decreased. Conclusion: Ckip-1 may affect auricular development in mice via regulating the proliferation and differentiation of ehondrocytes.
