摘 要:目的:探究大鼠腮腺萎缩再生过程的规律。方法:结扎198只Wistar大鼠保侧腮腺主导管7(A组)、14(B组)、21 d(C组)(n=66)后分别实现导管再通,采用HE染色、免疫组织化学染色和Tunel荧光检测观察再通后第0、1、3、5、7、10、14、21、28、60、90 d(记为A_0……A_(90)、B_0……B_(90)、C_0……C_(90),n=6)腮腺组织萎缩再生规律。另取6只鼠不结扎为对照组。结果:导管再通第0天,腺泡萎缩,导管扩张,管腔增大,PCNA、P53表达量以及细胞凋亡比例较正常对照组高,随后逐渐增多,并于A3、B5天达到最高值,后逐步减少,至A14、B21接近正常对照组水平,腺体组织结构恢复正常,而C组未见明显恢复。结论:腮腺导管结扎7、14 d后实现再通,腺体可完全恢复;结扎21 d后,则不能完全恢复甚至完全不能恢复。Objective: To explore the rule of the rat parotid gland regeneration after obstructive atrophy. Methods: The parotid ducts on right side of 198 Wistar rats were ligated for 7 d( group A) , 14 d( group B) 21 d( group C) ( n = 66) respectively and then released. 6 rats without duct ligation were used as the controls. HE staining, Immunohistoehemieal and Tune1 fluorescence detection were used to observe cell apoptosis and tissue regeneration of parotid specimens which were obtained at 0, 1, 3, 5, 7, 10, 14, 21, 28, 60 and 90 d respectively after releasing the duct( marked as A0 ……A90, B0 ……B90, Co…… C90, n = 6). Results: At day 0 after duct release, acinar cell atrophy and duct ectasia were observed. The expression of PCNA and P53 and cell apoptotie ratio were higher than those of the controls, and increased gradually to the highest value in A3, B5, then decreased gradually to those of the controls in A14 and B21. In A14 and B21 the gland tissue structure returned to normal. No any regeneration manifestation was observed in group C. Condusion: After release of the ligated duct, the parotid gland can return to normal within the appropriate time, otherwise, it will be atrophied and can not be recovered.
