摘 要:以收集引进的40个野生香菇菌株作为试验材料,通过酯酶同工酶技术和ISSR分子标记技术,对其遗传多样性进行分析。结果表明:酯酶同工酶酶谱显示野生香菇菌株不同迁移率的酶带多达15条,聚类分析表明,当遗传相似系数约达71%时,将40个野生香菇菌株分为七大类群;通过ISSR分子标记共扩增出75条清晰的DNA多态片段,大小介于0.2~4.0kb;聚类分析表明,在遗传相似系数约达63%时,将40个野生香菇菌株分为七大类群。该研究为野生香菇种质资源驯化奠定了一定的基础。Forty wild Lentinus edodes strains were used as experimental materials,the genetic diversity were identified and analyzed by esterase isoenzyme zymogram and ISSR molecular marker technique.The results showed that fifteen distinct bands were detected by the esterase isozyme zymogram.Cluster analysis results showed the forty wild Letinous edodes strains were divided into seven groups at genetic similarity of 71%.Seventy-five distinct bands which ranged from 0.2 kb and 4.0 kb were detected by gel electrophoresis.Cluster analysis results showed the forty wild Letinous edodes strains were divided into seven groups at genetic similarity of 63%.This study had laid a foundation for the research on germplasm resources of wild strains of Lentinus edodes and breed domestication.
