摘 要:目的:建立同时测定中药及保健品中添加的9种改善肠胃功能类化学药物的超高效液相色谱-串联质谱(UPLC-MS/MS)法。方法:采用Agilent ZORBAX SB-C18(2.1 mm×50 mm,1.8μm)色谱柱,以乙腈-0.1%甲酸溶液(22∶78)为流动相,流速为0.2 m L/min。质谱采用电喷雾离子源(ESI)正离子检测,以多重反应监测(MRM)方式进行定量。结果:9种药物在10~1,000 ng/m L范围内呈现良好的线性关系,r>0.998。9种药物在10 ng/m L添加水平的加样回收率93.6%~102.4%,RSD%<5%。15批样品中检出3批阳性样品。结论:该方法可作为中药及保健品中非法添加西咪替丁、雷尼替丁、法莫替丁、奥美拉唑、泮托拉唑、兰索拉唑、多潘立酮、莫沙必利及西沙必利等9种化学药物的检测方法。Objective: To establish a UPLC-MS/MS method for the determination of 9 supplementary chemical drugs traditional Chinese medicines and health care products for improving gastrointestinal function. Methods:The analysis was performed on a Agilent ZORBAX SB-C18 column(2.1 mm×50 mm, 1.8 μm). Mobile phase was acetonitrile-0.1% formic acid water solution(22 ∶78) at a flow rate of 0.2 m L/min. Electrospray ionization(ESI)source was applied in the positive mode. Multiple reaction monitoring(MRM) mode was used to quantify 9 chemical drugs. Results: The correlation coefficients of 9 drugs were all more than 0.998 with linear range 10-1,000 ng/m L. The average recovery of 9 drugs at the spiked level of 10 ng/m L was between 93.6%-102.4% with the relative standard deviation less than 5%. Three positive samples in 15 batchs samples were detected. Conclusion: The method can be used to determin 9 chemical drugs, including cimitidine, ranitidine, famotidine,omeprazole, pantoprazole, lansoprazole, domperidone, mosapride and cisapride.
