摘  要:目的:观察脊柱推拿对脑性瘫痪模型幼鼠学习记忆的影响,并探索其机制。方法:将40只雄性幼鼠随机分为空白对照组、正常推拿组、模型对照组、模型推拿组,每组10只。模型对照组、模型推拿组在出生后第3天制备脑瘫模型,并于出生后第5天开始进行脊柱推拿,持续28 d。采用Morris水迷宫实验和跳台实验观察幼鼠学习记忆能力,采用高效液相色谱法检测大鼠海马组织内5-羟色胺、多巴胺、肾上腺素、去甲肾上腺素含量。结果:模型对照组和模型推拿组在Morris水迷宫实验中找到平台的潜伏期高于空白对照组;正常推拿组穿越平台的次数多于空白对照组(P<0.05);跳台实验中,模型对照组跳下平台的潜伏期明显低于空白对照组,跳下平台的次数明显多于空白对照组(P<0.05)。模型对照组海马组织内去甲肾上腺素和5-羟色胺含量明显低于空白对照组(P<0.05);正常推拿组、模型推拿组海马组织内肾上腺素均明显高于空白对照组(P<0.05);正常推拿组和模型推拿组肾上腺素明显高于模型对照组,差异有统计学意义(P<0.05)。模型对照组海马组织内多巴胺明显高于空白对照组,差异有统计学意义(P<0.05)。结论:脊柱推拿可改善脑瘫模型幼鼠学习记忆能力,可能与调节海马内5-羟色胺、去甲肾上腺素、肾上腺素、多巴胺等单胺类神经递质有关。Objectives: To observe the effect of Spinal Tuina on the learning and memory in the hippocampus of cerebral palsy(CP) juvenile rats, and explore the mechanism. Methods: A total of 40 male juvenile rats were randomly divided into blank control group, control+Tuina group, model group and model+Tuina group, with10 rats in each. The model group and the model+Tuina group were operated to establish CP model in postnatal3 days(P3). After P5, the control and Tuina group and model and Tuina group were given spinal Tuina for 28 d. Morris water maze(MWM) and Step-down test(ST) were commenced respectively to observe learning and memory abilities. The levels of 5-hydroxytryptamine(5-HT), norepinephrine(NE), adrenaline(E) and dopamine(DA) in hippocampus tissue were detected by high performance liquid chromatography. Results: The model group and the model+Tuina group showed longer incubation of finding the platform than the blank control group in the MWM, and the control +Tuina group showed higher number of crossing the platform than blank control group(P〈0.05). The model group showed short incubation period than the blank control group as well as more times of jumping down the platform in the ST(P〈0.05). The model group showed lower content of NE and 5-HT hippocampus tissue than the blank control group(P〈0.05). The ontrol+Tuina and model+Tuina group showed higher content of adrenaline than the blank control group(P〈0.05). The control+Tuina and model+Tuina group showed higher content of adrenaline than the model group(P〈0.05). The model group showed higher content of DA than the blank control group(P〈0.05). Conclusion: Spinal Tuina can improve the learning and memory ability of juvenile rats with cerebral palsy. It may be related to the regulation of monoamine neurotransmitters such as 5-HT,NE, E and DA in the hippocampus.
