摘 要:目的探究石甘散对戊四氮(PTZ)致痫大鼠海马组织内谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平的影响。方法雌性SD大鼠120只,随机分为空白组、模型组、西药对照组、石菖蒲组、甘松组、石甘散组,每组20只。首先将实验大鼠适应性饲养7 d后,除空白组予腹腔注射0.9%氯化钠溶液1 ml外,各组均腹腔注射35 mg/kg PTZ进行造模。各组大鼠在造模完成1 d后开始灌胃给药,共治疗28 d。治疗结束,观察比较各组大鼠行为学[痫性发作潜伏期(ESL)和强直性惊厥潜伏期(TCL)]表现,计算惊厥发生率(TSR)。完成行为学监测后取材,观测大鼠海马组织内GSH-Px、MDA、SOD的含量。结果与模型组相比,各组均能延长ESL及TCL(P<0.05),且西药对照组、石甘散组明显长于石菖蒲组及甘松组(P<0.05)。各组TSR均低于模型组的100%,且西药对照组及石甘散组的TSR下降明显(P<0.05)。与模型组相比,各给药组均可以提高大鼠海马组织内SOD及GSH-Px活性,降低MDA含量(P<0.05);其中,西药对照组及石甘散组的结果均优于石菖蒲组、甘松组,差异有统计学意义(P<0.05),西药对照组与石甘散组相比,差异无统计学意义(P>0.05)。结论石甘散可以延长PTZ致痫大鼠的ESL及TCL,降低TSR。可通过提高海马组织内SOD及GSH-Px活性,降低MDA的含量,抑制氧化反应,减轻自由基对脑组织的损伤。Objective To explore the effects of shigansan on the content of GSH-Px,MDA and SOD in hippocampal tissue of the epileptic rats induced by pentrazole. Methods A total of 120 SD female rats were randomized into a blank group,a model group,a western medicine control group,a rhizome acori graminel group,a rhizome nardostachyos group and a shigansan group,20 rats in each one. After 7 d adaption feeding,except the blank group in which 0. 9% Na Cl1 ml was injected intraperitoneally,the modeling was done with intraperitoneal injection,35 mg/kg PTZ in the rest groups.The medication was given individually in each group through lavage since the 1 d after successful modeling in each group.Totally,the treatment lasted 28 d. At the end of treatment,the behavior manifestations(the epileptic seizure latency(ESL)and the tonic convulsion latency(TCL)) were compared among the groups. The tonic seizure rate(TSR) was calculated. The tissues were collected after behavior monitoring. The contents of GSH-Px,MDA and SOD in hippocampal tissues were observed. Results Compared with the model group,ESL and TCL were extended in each group(P 〈 0. 05). The extension was more obvious in the western medicine control group and the shigansan group as compared with rhizome acori graminel group and rhizome nardostachyos group(P 〈 0. 05). TSR in each group was lower than the model group and it was reduced remarkably in the western medicine control group and the shigansan group(P 〈 0. 05). Compared with the model group,SOD and GSH-Px activity in the hippocampal tissue were improved and MDA decreased in each medicine group(P〈 0. 05). In which,the results in the western medicine control group and the shigansan group were better than rhizome acori graminel group and rhizome nardostachyos group,indicating the significant differences(P 〈 0. 05). The differences were not significant between the western medicine group and the shigansan group(P〈0.05).Conclusion Shigansan prolongs ESL and TCL of PTZ induced epilepsy in the rats
