玉米基因ZmSGL增加水稻粒长和粒重功能的初步研究

摘  要:从玉米中克隆了一个基因ZmSGL(Zea mays L.stress tolerance and grain length),该基因与水稻粒长基因OsSGL的编码序列一致性为62.3%。本工作在增加水稻粒长和粒重的功能方面对ZmSGL进行了初步研究。研究表明,ZmSGL编码推定的DUF1645超家族蛋白。在水稻中表达玉米ZmSGL基因可引起水稻种子极显著增长增重。与野生型水稻相比,转基因水稻粒长增长10.6%,千粒重增重8.4%。转基因水稻小穗颖壳中的细胞数量和细胞大小增加,幼穗中CDKA1、CYCA2;1和CYCB2;1等细胞周期基因的表达水平升高。玉米基因ZmSGL作为谷粒大小调节因子,可应用于改善主要农作物的谷粒性状。Gene ZmSGL(Zea mays L. stress tolerance and grain length) was cloned from Zea mays, and the coding sequence of ZmSGL shares 62.3% homology with the rice grain length gene OsSGL. In our study, the function of ZmSGL in increasing grain length and weight of rice was investigated. The results showed that ZmSGL encoded a putative DUF1645 superfamily protein. The grain length and weight of transgenic rice were significantly increased with the expression of ZmSGL. Compared with WT(wild-type rice), the grain length of transgenic rice was increased by 10.6%, and 1 000-grain weight was increased by 8.4%. The number and size of cells in the spikelet of transgenic rice were increased, and the expression levels of CDKA1, CYCA2;1, CYCB2;1 and other cell cycle genes were increased in young panicles. As a regulator of grain size, maize gene ZmSGL could be used to improve grain traits in staple crops.
