CISH基因多态性与河北地区肺结核易感性的相关性研究

摘  要:目的探讨细胞因子诱导含SH2结构域蛋白(cytokine-inducible SH2-containing protein,CISH)基因单核苷酸多态性(Single nucleotide polymorphism,SNP)在河北地区汉族人群中的分布及其与肺结核(Tuberculosis,TB)易感性的关系。方法利用SNa Pshot技术分析法检测河北地区205名健康人(汉族)和214例肺结核病人(汉族)CISH基因3个位点rs414171、rs2239751和rs6768300位点的多态性,并进行基因型频率和等位基因频率的比较,通过统计学方法分析这些基因位点的多态性与肺结核易感性之间的相关性。结果选取的214例肺结核病人和205名正常人的年龄(t=0.014,P>0.05)、性别(χ~2=0.021,P>0.05)差异均无统计学意义,rs414171、rs2239751和rs6768300位点的基因频率分布均符合Hardy-Weinberg平衡定律(P>0.05)。与对照组相比,结核组的rs414171位点的基因型(TT、TA、AA)(χ~2=4.052,P>0.05)、等位基因(T/A)(χ~2=2.189,P>0.05),rs2239751位点的基因型(AA、AC、CC)(χ~2=1.265,P>0.05)、等位基因(A/C)(χ~2=1.177,P>0.05),rs6768300位点的基因型(AA、AC、CC)(χ~2=0.908,P>0.05)、等位基因(A/C)(χ~2=0.729,P>0.05)差异均无统计学意义。结论 CISH基因的rs414171、rs2239751和rs6768300位点多态性与河北地区汉族人群的肺结核易感性没有显著的相关性。Objective To explore the association between single nucleotide polymorphisms(SNP) in cytokine-inducible SH2-containing protein gene and the susceptibility to tuberculosis(TB). Methods 205 cases of normal people and 214 cases of tuberculosis patients(both of which are from the han nationality) were analyzed by the method of micro-sequencing technology(SNa Pshot) in Hebei via the genotypic and allelic frequencies of CISH(rs414171,rs2239751 and rs6768300). The relationship between CISH genotypes and the susceptibility to tuberculosis was analyzed by logistic regression models. Results There was no significant difference between the age(t = 0. 014,P〈0. 05) and gender(χ2= 0. 021,P〈0. 05) of 214 patients with tuberculosis and 205 cases of normal people. The distribution of gene frequency in rs414171,rs2239751 and rs6768300 conformed to the Hardy-Weinberg equilibrium law by χ2 test(P〈0. 05). Compared with control group,there was no significant difference on the rs414171 genotype(TT,TA and AA,χ2= 4. 052,P〈0. 05) and allelic frequencies(T/A,χ2= 2. 189,P〈0. 05),the genotype of rs2239751 loci(AA,AC and CC,χ2= 1. 265,P〈0. 05) and allele(A/C,χ2= 1. 177,P〈0. 05),and the genotype of rs6768300 loci(AA, AC and CC, χ2= 0. 908, P〈0. 05) and allele(A/C, χ2= 0. 729, P〈0. 05) of TB group. Conclusion The polymorphism of CISH(rs414171,rs2239751 and rs6768300) has no significant correlation with the incidence of tuberculosis of people of the han nationality in hebei province.
