摘  要:对P0、P7、P14、P30小鼠的大脑组织切片进行DNA甲基转移酶1(Dnmt1)、Reelin免疫荧光标记,在荧光显微镜下观察小鼠大脑皮质Cajal-Retzius(CR)细胞中Reelin与Dnmt1阳性细胞分布与密度变化,探讨小鼠大脑新皮质片层化发育过程中CR细胞DNA甲基化的改变以及对新皮质发育的影响.结果表明,Reelin-Dnmt1双标记阳性细胞、Reelin阳性细胞密度随大脑皮质的发育逐渐降低,但是Reelin-Dnmt1双标记阳性细胞占Reelin阳性细胞的比例先升高,并在14d日龄时达到最大值,然后逐渐降低.提示随着大脑皮质的发育,CR细胞DNA甲基化状态发生改变,CR细胞DNA甲基化在神经元的迁移与定位中发挥重要作用.Immunofluorescent double labeling was performed to investigate the expression and distribution of Reelin,Dnmt1 in Cajal-Retzius(CR)cells in neocortex from P〈0,P7,P14,and P30 mice.The changes of DNA methylation in CR cells and the effects on neocortex development in neoplastic cortical development of mouse brain were explored.The results show that the density of Reelin-Dnmt1 double-labelled positive cells and Reelin-positive cells gradually decreases with the development of cerebral cortex,but the proportion of Reelin-Dnmt1 double-labelled positive cells in Reelin-positive cells increases first and reaches a maximum at 14 dof age,and then gradually lowers.It is suggested that with the development of cerebral cortex,the DNA methylation status of CR cells changes,and DNA methylation of CR cells plays an important role in the migration and localization of neurons.
