华中冬青雌雄株抗寒生理指标筛选及评价  被引量:14

摘  要:以华中冬青离体叶片为供试材料,通过低温胁迫处理,测定其相对电导率(Rec)、丙二醛(MDA)、可溶性蛋白(SP)、可溶性糖(SS)、游离脯氨酸(Pro)、超氧化物歧化酶(SOD)、叶绿素(Chl)和类胡萝卜素(Car)8项生理指标,结合隶属函数值和半致死温度对其抗寒性进行综合评价,筛选适合评价华中冬青的抗寒性生理指标,比较华中冬青雌雄株的抗寒性。结果表明:低温胁迫下华中冬青的Rec和MDA含量随着温度的降低呈"S"形曲线上升,SP含量、Pro含量和SOD活性随着温度的降低呈先上升后下降的趋势,Chl含量随着温度的降低而降低,Car随着温度的降低基本不变;对8个抗寒指标进行相关性分析和主成分筛选发现,低温胁迫与Rec和MDA呈极显著负相关,与SP和Chl呈显著正相关;Rec、MDA、SP、SOD和Chl含量变化与华中冬青的抗寒性关系密切,可作为评价其抗寒性的指标;华中冬青雌雄株的半致死温度分别为0.544℃(雌株)和1.027℃(雄株),雌雄株的抗寒性差异不显著,雌株抗寒性略强于雄株,与综合分析结果一致。The isolated leaves of the male and female strains of Ilex centrochinensis were used as test materials, through low temperature stress to determine the relative electric conductivity(Rec), malondialdehyde(MDA), soluble protein(SP) and soluble sugar(SS), free proline(Pro), superoxide dismutase(SOD), chlorophyll(Chl) and carotenoidin(Car) 8 physiological indexes, combined with the value of comprehensive evaluation of cold resistance physiological indexes for the screening of membership function, evaluation of cold resistance of I. centrochinensis. The results showed that Rec and MDA content under low temperature stress of I. centrochinensis with decreasing temperature is "S" shaped curve rose, SP content, Pro content and SOD activity decreased with the temperature increased first and then decreased, Chl content decreased with the decrease of temperature. Correlation analysis and principal component analysis of 8 cold resistant indexes showed that the low temperature stress had a significant negative correlation with Rec and MDA, and was positively correlated with SP and Chl; The changes of Rec, MDA, SP, SOD and Chl were closely related to the cold resistance of I. centrochinensis, and could be used as an index to evaluate its cold resistance. The semi lethal temperature of male and female plants in I. centrochinensis was calculated by Logistic equation, which was 1.027 ℃(male plant) and 0.544 ℃(female strain). There was no significant difference in the cold resistance of the female and male plants, but the female plants were slightly stronger than the male plants, which was consistent with the results of the comprehensive analysis.
