住院患者伏立康唑与伊马替尼联合应用的稳态药动学研究

摘　　要：目的:研究住院患者联合应用伏立康唑与伊马替尼对药物谷浓度及稳态药动学的影响。方法:采用高效液相色谱法同时测定住院患者联合应用伏立康唑前后伊马替尼与伏立康唑的稳态血浆药物浓度,采用DAS 3.2.8程序计算出药动学参数,采用SAS 9.0软件对谷浓度和主要药动学参数进行统计分析。结果:联合用药后,伊马替尼的谷浓度较联合用药前明显升高,差异有统计学意义(P<0.05);伏立康唑的谷浓度高于有效浓度;而联合用药前后伊马替尼主要药动学参数之间的差异均无统计学意义(P>0.05)。结论:伊马替尼与伏立康唑联合应用可升高伊马替尼的谷浓度,而对其药动学影响不大,联合用药期间宜监测伏立康唑的血药浓度。OBJECTIVE： To probe into the effects of voriconazole combined with imatinib on drug valley concentration and steady-state pharmacokinetics in inpatients. METHODS： HPLC method was adopted to determine the drug valley concentration and steady-state pharmacokinetics in inpatients before and after application of voriconazole combined with imatinib, DAS 3.2. 8 was used to calculate the pharmacokinetic parameters, SAS 9.0 software was adopted to statistically analyze the drug valley concentration and main pharmacokinetics parameters. RESULTS： After drug combination, the drug valley concentration of imatinib increased significantly compared with before drug combination, with statistically significant difference （P 〈 0.05 ）. The drug valley concentration of voriconazole was higher than the effective concentration. Yet there was no statistical significance between two groups in the pharmacokinetic parameters of imatinib before and after drug combination（P 〉 0.05）. CONCLUSIONS ： Voriconazole combined with imatinib can increase the drug valley concentration of imatinib, yet has few influence on the pharmacokinetic. And the blood concentration of voriconazole should be monitored during drug combination.
